6. Mapping the human proteome using bioinformatic methods
Sammanfattning : The fundamental goal of proteomics is to gain an understanding of the expression and function of the proteome on the level of individual proteins, on the level of defined cell types and on the level of the entire organism. In this thesis, the human proteome is explored using membrane protein topology prediction methods to define the human membrane proteome and by global protein expression profiling, which relies on a complex study of the location and expression levels of proteins in tissues and cells. 7. Targeted proteomics methods for protein quantification of human cells, tissues and blood
Sammanfattning : The common concept in this thesis was to adapt and develop quantitative mass spectrometric assays focusing on reagents originating from the Human Protein Atlas project to quantify proteins in human cell lines, tissues and blood. The work is based around stable isotope labeled protein fragment standards that each represent a small part of a human protein-coding gene. LÄS MER
8. Antibody-based bead arrays for high-throughput protein profiling in human plasma and serum
Sammanfattning : Affinity-based proteomics utilizes affinity binders to detect target proteins in a large-scale manner. This thesis describes a high-throughput method, which enables the search for biomarker candidates in human plasma and serum. LÄS MER
9. The physiological processing of Alzheimer-associated amyloid beta precursor protein in human and animal-derived neuronal models
Sammanfattning : Alzheimer’s disease (AD) is characterized by cognitive impairment due to the loss of structure and/or function of neurons, and amyloid plaques composed of aggregated-amyloid beta (Aβ) peptides, primarily species ending at the amino acid 42 (Aβ42), are one of the major neuropathological hallmarks of AD. Aβ peptides of different lengths are produced by sequential cleavage of amyloid beta precursor protein (APP) by α-, β- and γ- secretases. LÄS MER
10. Interactions between Streptococcus pyogenes and the human immune defence
Sammanfattning : Streptococcus pyogenes is an important human pathogen frequently colonizing the throat and skin of humans. To facilitate colonization and spread and to avoid the host immune defence, streptococci are endowed with a variety of virulence factors, two of which are investigated in this thesis namely streptolysin O (SLO) and M protein.
