  1. 26. On the binding of growth-promoting polyamines to proteoglycans: Implications for growth-regulation and polycation-mediated gene transfer

    Författare :Mattias Belting; Institutionen för experimentell medicinsk vetenskap; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Genetics; cationic lipid.; gene transfer; gene therapy; cell growth; polyamine transport; spermine; polyamine; growth-inhibition; heparan sulphate; dermatan sulphate; Proteoglycan; glycosaminoglycan; cytogenetics; Genetik; cytogenetik;

    Sammanfattning : Initial investigations were directed at studying the interaction between polyamines and various glycosaminoglycans (GAGs). The polyamine spermine displayed binding to dermatan sulphate (DS) and heparan sulphate (HS) with similar (Kd, 3.9 x 10-4 M) and higher (Kd, 0.37 x 10-6 M) affinity, respectively, than to DNA. 2. 27. From Genes to Therapy : Modeling and Novel Therapeutic Strategies for Brain Tumors

    Författare :Matko Čančer; Fredrik Swartling; Lene Uhrbom; Suzanne J. Baker; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Medulloblastoma; Glioblastoma; MYCN; PDGF; Cancer Therapy; BET bromodomains; Temozolomide; Medicinsk vetenskap; Medical Science; Oncology; Onkologi;

    Sammanfattning : Medulloblastoma is the most common malignant pediatric brain tumor and is molecularly divided into four subgroups – WNT, SHH, Group 3 and Group 4. Two thirds of medulloblastoma patients survive, but survivors often suffer from severe, lifelong side-effects. The MYCN oncogene is deregulated in many medulloblastoma patients. LÄS MER

  3. 28. CD40L Gene Therapy for Solid Tumors

    Författare :Lina Liljenfeldt; Thomas Tötterman; Angelica Loskog; Sara Mangsbo; Evren Alici; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; CD40L; adenovirus; tumor immunology; tumor microenvironment; immunotherapy; local immunotherapy; antigen presentation; Immunologi; Immunology;

    Sammanfattning : Adenoviral CD40L gene therapy (AdCD40L) is a strong inducer of anti-tumor immune responses via its activation of dendritic cells (DCs). Activated DCs can in turn activate T cells, which are key players in an efficient anti-tumor response.This thesis includes three papers that focus on different aspects of AdCD40L gene therapy. 4. 29. Development of lentiviral vectors for CNS gene transfer

    Författare :Johan Jakobsson; Brain Repair and Imaging in Neural Systems (BRAINS); []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Medicin människa och djur ; Medicine human and vertebrates ; Biotechnology; Gene Therapy; Neurological disorders;

    Sammanfattning : Gene therapy in the brain is a promising treatment strategy that in the future may be used for several brain disorders, including Parkinson's disease, Alzheimer's disease and lysosomal storage diseases. By introducing a new gene, rather than providing a classical pharmacological drug, gene therapy offers the opportunity to treat neurological disorders by a single intervention. LÄS MER

  5. 30. Mechanisms of Transgene Silencing in Neural Cells -Implications for Ex Vivo Gene Therapy to the Brain

    Författare :Nina Rosenqvist; Neurobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neurophysiology; Neurologi; Neurology; neuropsychology; ChIP; rat; transgene; insulator; differentiation; GFP; lentivirus; DNA methylation; histone deacetylation; chromatin; expression; silencing; stem cell; progenitor; gene therapy; neurofysiologi; neuropsykologi; CNS; transplantation;

    Sammanfattning : The use of genetically modifi ed neural stem and progenitor cells could possibly be a future strategy for repairing a brain suffering from neurodegeneration. However, so far it has proven diffi cult to maintain sufficient levels of a transferred gene in suitable cell types, after transplantation. LÄS MER
