11. THE ROLE OF PHOSPHODIESTERASE 3B IN CAMP-MEDIATED REGULATION OF INSULIN SECRETION
Sammanfattning : Type 2 diabetes mellitus (T2DM) is characterized by various combinations of ?-cell failure and insulin resistance leading to hyperglycemia and glucose intolerance. In order to maintain glucose tolerance in the insulin resistance state, increased insulin secretion is a requirement and it is because of inadequate islet adaptation that glucose intolerance develops in T2DM. 12. Proinsulin and Insulin Sensitivity as Predictors of Type 2 Diabetes Mellitus and Coronary Heart Disease : Clinical Epidemiological Studies with up to 27 Years of Follow-Up
Sammanfattning : Defects in insulin secretion and insulin action are the major abnormalities in the development of Type 2 diabetes. Hyperinsulinemia is a risk marker for Type 2 diabetes and according to some, but not in all studies also for coronary heart disease (CHD). Conventional insulin assays measure immunoreactive insulin including proinsulin-like molecules. LÄS MER
13. On Cholecystokinin and Phospholipase A2 in Insulin Secretion - an Experimental Study With Reference to the Development of Diabetes
Sammanfattning : In view of unsolved issues regarding cellular events underlying the adaptation of insulin secretion to insulin resistance, together with the observation that insulin secretion induced by the gastrointestinal hormone and neuropeptide cholecystokinin (CCK) is not solely explained by phospholipase C-protein kinase C (PLC-PKC) pathway activation, this study aimed at delineate mechanisms responsible for the hyperinsulinemia in insulin resistance, with particular attention paid to the action of glucose vs. non-glucose secretagogues, such as CCK. 14. Mechanisms by which variants in the TCF7L2 gene increase the risk of developing Type 2 diabetes
Sammanfattning : Type 2 diabetes mellitus (T2DM) is a heterogeneous disease with a multifactorial aetiology comprising of genetic and environmental factors. The common variant most highly associated with T2DM known to date is a SNP rs7903146 in the TCF7L2 gene. However, the role TCF7L2 plays in the development of T2DM was unclear. LÄS MER
15. Pharmacometrics Modelling in Type 2 Diabetes Mellitus : Implications on Study Design and Diabetes Disease Progression
Sammanfattning : Pharmacometric modelling is widely used in many aspects related to type 2 diabetes mellitus (T2DM), for instance in the anti-diabetes drug development, and in quantifying the disease progression of T2DM.The aim of this thesis were to improve the design of early phase anti-diabetes drug development studies with the focus on the power to identify mechanism of drug action (MoA), and to characterize and quantify the progression from prediabetes to overt diabetes, both the natural progression and the progression with diet and exercise interventions, using pharmacometrics modelling.
