1. Mechanisms by which variants in the TCF7L2 gene increase the risk of developing Type 2 diabetes
Sammanfattning : Type 2 diabetes mellitus (T2DM) is a heterogeneous disease with a multifactorial aetiology comprising of genetic and environmental factors. The common variant most highly associated with T2DM known to date is a SNP rs7903146 in the TCF7L2 gene. However, the role TCF7L2 plays in the development of T2DM was unclear. LÄS MER
2. Gene x lifestyle interactions in type 2 diabetes mellitus and related traits
Sammanfattning : Background: Type 2 diabetes is thought to result from interactions between genetic and lifestyle factors, but few robust examples exist. The overarching aim of this thesis was to discover such interactions by studying cohorts of white youth and adults from northern Europe in which physical activity, genotypes, and diabetes-related traits or diabetes incidence had been ascertained. 3. Identification of susceptibility genes in type 2 diabetes
Sammanfattning : Identification of the susceptibility genes will offer better understanding of molecular mechanisms underlying T2D pathogenesis, and subsequently may lead to development of novel therapeutic approaches. This thesis mainly concerns the genetic association study of four candidate genes. LÄS MER
4. The genetic background of gestational diabetes mellitus
Sammanfattning : The aims of this work is to better determine the genetic background of gestational diabetes (GDM) and to examine how specific genes affect the development of diabetes post partum. In the DiPiS (Diabetes Prediction in Skåne) study we typed for HLA-DQB1 alleled, the transcription factor 7-like 2(TCF7L2) rs7903146, rs12255372 and rs7901695 SNPs, the 1858 C>T SNP of the protein tyrosine phosphatase non-receptor 22 (PTPN22) gene and we tested for the presence of islet cell autoantibodies against glutamic acid decarboxylase 65 (GAD65), insulinoma-associated antigen 2 (IA-2) and insulin in women who had been diagnosed with GDM at least once during 2000-2004. 5. Diabetes mellitus and risk of prostate cancer - A focus on shared risk factors and survivor bias
Sammanfattning : Prostate cancer (PCa) is a heterogenous disease including very aggressive lethal tumors to incidentally discovered clinically insignificant tumors. Age, ethnicity, and family history are the only established risk factors. LÄS MER
