Sammanfattning : ABSTRACT Low oxygen tension (hypoxia) is a major inducer of neovascularisation and evidence emerging has indicated that oxygen tension and hypoxia-inducible factors (HIFs), a family of oxygen-regulated heterodimeric transcription factors, may play important roles in vascular and hematopoietic development. This thesis has focused on studying molecular and cellular mechanisms, by which cells and organisms sense changes in oxygen tension, and how this signal is transduced by HIFs into downstream signalling. Sammanfattning : In the present thesis, the quantitative role of active transport (transcytosis) in bulk transvascular protein movement in vivo was evaluated. Using chemical transcytosis inhibition (N-ethylmaleimide (NEM) and filipin), we were able to show that NEM actually increases endothelial permeability to macromolecules, probably as a result of unspecific toxic action on endothelial cells. PSA occurs in several molecular forms in blood: mainly as a free form with a molecular mass of 28 kDa; and as PSA complexed to alpha-1-antichymotrypsin, complexed PSA, with a molecular mass of 90 kDa.
