26. On the binding of growth-promoting polyamines to proteoglycans: Implications for growth-regulation and polycation-mediated gene transfer
Sammanfattning : Initial investigations were directed at studying the interaction between polyamines and various glycosaminoglycans (GAGs). The polyamine spermine displayed binding to dermatan sulphate (DS) and heparan sulphate (HS) with similar (Kd, 3.9 x 10-4 M) and higher (Kd, 0.37 x 10-6 M) affinity, respectively, than to DNA. 27. From Genes to Therapy : Modeling and Novel Therapeutic Strategies for Brain Tumors
Sammanfattning : Medulloblastoma is the most common malignant pediatric brain tumor and is molecularly divided into four subgroups – WNT, SHH, Group 3 and Group 4. Two thirds of medulloblastoma patients survive, but survivors often suffer from severe, lifelong side-effects. The MYCN oncogene is deregulated in many medulloblastoma patients. LÄS MER
28. CD40L Gene Therapy for Solid Tumors
Sammanfattning : Adenoviral CD40L gene therapy (AdCD40L) is a strong inducer of anti-tumor immune responses via its activation of dendritic cells (DCs). Activated DCs can in turn activate T cells, which are key players in an efficient anti-tumor response. This thesis includes three papers that focus on different aspects of AdCD40L gene therapy. 29. Development of lentiviral vectors for CNS gene transfer
Sammanfattning : Gene therapy in the brain is a promising treatment strategy that in the future may be used for several brain disorders, including Parkinson's disease, Alzheimer's disease and lysosomal storage diseases. By introducing a new gene, rather than providing a classical pharmacological drug, gene therapy offers the opportunity to treat neurological disorders by a single intervention. LÄS MER
30. Mechanisms of Transgene Silencing in Neural Cells -Implications for Ex Vivo Gene Therapy to the Brain
Sammanfattning : The use of genetically modifi ed neural stem and progenitor cells could possibly be a future strategy for repairing a brain suffering from neurodegeneration. However, so far it has proven diffi cult to maintain sufficient levels of a transferred gene in suitable cell types, after transplantation. LÄS MER
