  1. 1. Mechanisms of Transgene Silencing in Neural Cells -Implications for Ex Vivo Gene Therapy to the Brain

    Författare :Nina Rosenqvist; Neurobiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neurophysiology; Neurologi; Neurology; neuropsychology; ChIP; rat; transgene; insulator; differentiation; GFP; lentivirus; DNA methylation; histone deacetylation; chromatin; expression; silencing; stem cell; progenitor; gene therapy; neurofysiologi; neuropsykologi; CNS; transplantation;

    Sammanfattning : The use of genetically modifi ed neural stem and progenitor cells could possibly be a future strategy for repairing a brain suffering from neurodegeneration. However, so far it has proven diffi cult to maintain sufficient levels of a transferred gene in suitable cell types, after transplantation. LÄS MER

  2. 2. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors

    Författare :Niels-Bjarne Woods; Andreas Ooka; Stefan Karlsson; Avdelningen för molekylärmedicin och genterapi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; embryonic stem cell; Hematopoietic stem cell; lentiviral vector; transduction efficiency; transgene expression; multiple vector copy integration; insertional mutagenesis; NOD SCID mouse; cytogenetics; Genetics; extracellulära vätskor; extracellular fluids; cytogenetik; Genetik; Haematology; Hematologi; Lentivirus : genetics; Human; Genetic Vectors; Hematopoietic Stem Cells : metabolism; Animal; Gene Therapy; Leukemia : therapy; Leukemia : metabolism;

    Sammanfattning : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 3. 3. The Role of the SHB Adapter Protein in Cell Differentiation and Development

    Författare :Vitezslav Kriz; Michael Welsh; Urban Lendahl; Uppsala universitet; []
    Nyckelord :Cell biology; SHB; transgene; ES cells; cavitation; knockout; mouse; vasculogenesis; hematopoiesis; endoderm; mesoderm; malformation; Cellbiologi;

    Sammanfattning : The present study was conducted in order to assess a role of the SH2 domain-containing adapter protein SHB in development and cell differentiation.Embryonic stem (ES) cells overexpressing SHB and SHB with an inactive SH2 domain (R522K-SHB) were obtained. LÄS MER

  4. 4. Expression of Ig genes. Regulation of transcription and production of human antibodies

    Författare :Christina Furebring; Institutionen för immunteknologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Immunology; Bioteknik; Biotechnology; human antibodies; immunoglobulin gene expression; 3 enhancer; serology; transplantation; Immunologi; serologi;

    Sammanfattning : During B lymphocyte development, the transcriptional activity of the IgH locus is subject to spatial and temporal changes. The 3' enhancer (3'E) has been suggested to play an important role in regulation of immunoglobulin gene expression late in B cell development. LÄS MER

  5. 5. Expression and function of ribozymes in transgenic mice

    Författare :Michael Andäng; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :hammerhead ribozyme; hairpin ribozyme; transgenic mice; HIV-1; pseudotype virus; cleavage products; cis-cleavage;

    Sammanfattning : Hammerhead and hairpin ribozymes are short RNAs that act as sequence-specific endoribonucleases by cleaving a target RNA after hybridization to complementary sequences. By changing the specific target hybridization sequence of the ribozyme, virtually any RNA can be targeted, making ribozymes a versatile tool for gene down-regulation (knockdown). LÄS MER
