1. Development of Gene Therapy for Hematopoietic Stem Cells using Lentiviral Vectors
Sammanfattning : Hematopoietic stem cells are an ideal target for genetic manipulation for the purpose of curing hematological disorders as they have the ability to develop into all blood lineages and to self-renew. In this study we demonstrate that lentiviral vectors, based on HIV-1, can efficiently transfer genes into human hematopoietic progenitor and stem cells as assessed following stem cell transplantation in immune compromised mice. 2. RNA modifications and post-transcriptional control in cancer and stem cells
Sammanfattning : Splicing and translation are two of the key steps of post-transcriptional regulation of gene expression. Their tight regulation is essential for development, whereas their deregulation is involved in cancer pathogenesis. Nevertheless, many of the molecular mechanisms controlling these processes are still unknown. LÄS MER
3. Hypoxia inducible factor 1 alpha : dependent and independent regulation of hematopoietic stem cells and leukemia
Sammanfattning : This thesis has studied the role of low oxygen levels, or hypoxia, in hematopoietic stem cells (HSCs) and how, at the molecular level, it regulates stem cell maintenance and protects against oxidative stress induced by reactive oxygen species (ROS). HSCs reside within the bone marrow in specific niches created by a unique vascularized environment, which is suggested to be hypoxic and crucial for HSCs by maintaining a quiescent state of cell cycle and by redirecting metabolism away from the mitochondria to glycolysis. 4. Studies on arachidonic acid metabolism in normal and malignant hematopoietic cells
Sammanfattning : This thesis deals with the metabolism of arachidonic acid via the 5-lipoxygenase- and the 15-lipoxygenase-1 pathways in normal and malignant hematopoietic cells. The first part of the thesis (papers I & II) describes the expression of genes involved in arachidonic acid metabolism and the capacity to generate leukotriene (LT) B4 via the 5-lipoxygenase pathway in blood tumor cells from patients with precursor B-ALL (acute lymphoblastic leukemia) and AML (acute myeloid leukemia). 5. Laminins and Fibulins in Bone Marrow
Sammanfattning : Development of hematopoietic cells in bone marrow takes place in association with the hematopoietic microenvironment. Extracellular matrix (ECM) molecules and stromal cells in bone marrow provide a structural environment for hematopoietic cells, but they also directly influence proliferation, differentiation and migration of cells by signal transduction mediated by ligand binding to cell surface adhesion receptors.
