1. A multivariate approach to computational molecular biology
Sammanfattning : This thesis describes the application of multivariate methods in analyses of genomic DNA sequences, gene expression and protein synthesis, which represent each of the steps in the central dogma of biology. The recent finalisation of large sequencing projects has given us a definable core of genetic data and large-scale methods for the dynamic quantification of gene expression and protein synthesis. 2. Metabolic responsiveness to growth hormone in children
Sammanfattning : Metabolic effects of growth hormone (GH) therapy in short children have not been clearly established owing to the previous lack of controlled trials studying the metabolic outcome in response to different GH doses despite the known effects of GH on insulin sensitivity, lipid profile, and body composition. It has previously been shown that individualized GH doses during catch-up growth significantly reduce the proportion of unexpectedly good and poor responders around a predefined individual growth target in short prepubertal children. 3. Protein Profiling and Type 2 Diabetes
Sammanfattning : Type 2 diabetes mellitus (T2DM) is a heterogeneous disease affecting millions of people worldwide. Both genetic and environmental factors contribute to the pathogenesis. The disease is characterized by alterations in many genes and their products. LÄS MER
4. Helicobacter pylori cell surface interactions with glycosaminoglycans. Identification and characterisation of proteins binding to heparin/heparan sulphate
Sammanfattning : Helicobacter pylori is a gastric pathogen which cause chronic type B gastritis and peptic ulcer disease. H. pylori produces virulence factors such as urease, vacuolating cytotoxin VacA, cag pathogenicity island-associated proteins, flagella and adhesins. LÄS MER
5. Glucotoxicity in Insulin-Producing β-Cells
Sammanfattning : Background and aims: Type 2 diabetes mellitus is connected with elevated glucose levels, which cause impaired glucose-stimulated insulin secretion (GSIS) and degeneration of β-cells. Mechanisms for such glucotoxic effects were explored in the present study.Materials and methods: INS-1E cells were cultured for 5 days in 5. LÄS MER
