21. Developmental Aspects of Drug Transport Across the Blood-Brain Barrier
Sammanfattning : The developmental aspect of drug transport across the blood-brain barrier (BBB) was investigated. Microdialysis was used to study unbound morphine BBB transport at different ages in sheep. An in vitro study was performed to find differentially expressed genes in brain capillary-rich fractions of the brain in rats of different ages. LÄS MER
22. Development and Evaluation of Lipodisks Intended for Use as Biomimetic Membranes and Drug Carriers
Sammanfattning : Polyethylene glycol-stabilized lipodisks have emerged as a novel type of lipid-based nanoparticles with high potential as both drug carriers and biomimetic membranes. In this thesis we assess both of these applications, and show how the properties of the lipodisks can be further developed and optimized. LÄS MER
23. Preclinical and Clinical Development of the Novel Cyanoguanidine CHS 828 for Cancer Treatment
Sammanfattning : CHS 828 is a cyanoguanidine with anti-tumour properties which has shown promising effects in several preclinical models. This thesis describes both preclinical and clinical studies aiming to investigate disease specific activity, clinical tolerability and efficacy of CHS 828. LÄS MER
24. Development of Methods for Assessing Unbound Drug Exposure in the Brain : In vivo, in vitro and in silico
Sammanfattning : The blood-brain barrier is formed by tightly joined capillary cells with transporter proteins and acts as to regulate the brain concentration of nutrients as well as many drugs. When developing central nervous system drugs it is necessary to measure the unbound drug concentration in the brain, i.e. the unbound brain exposure. LÄS MER
25. Mixed Effects Modeling of Deterministic and Stochastic Dynamical Systems - Methods and Applications in Drug Development
Sammanfattning : Mathematical models based on ordinary differential equations (ODEs) are commonly used for describing the evolution of a system over time. In drug development, pharmacokinetic (PK) and pharmacodynamic (PD) models are used to characterize the exposure and effect of drugs. LÄS MER
