  1. 1. In vitro and in silico prediction of drug-drug interactions with transport proteins

    Detta är en avhandling från Uppsala : Acta Universitatis Upsaliensis

    Författare :Gustav Ahlin; Uppsala universitet.; [2009]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Solute carrier; SLC transporter; OCT1; Organic cation transporter 1; SLC22A1; OATP1B1; SLCO1B1; Organic anion transporting peptide 1B1; Drug transport; Active transport; Genetic polymorphism; Cell lines; Gene expression; Multivariate data analysis; OPLS; PHARMACY Pharmaceutics; FARMACI Galenisk farmaci; PHARMACY Biopharmacy; FARMACI Biofarmaci; galenisk farmaci; Pharmaceutics; Biopharmaceutics; biofarmaci;

    Sammanfattning : Drug transport across cells and cell membranes in the human body is crucial for the pharmacological effect of drugs. Active transport governed by transport proteins plays an important role in this process. A vast number of transport proteins with a wide tissue distribution have been identified during the last 15 years. LÄS MER

  2. 2. The Hepatobiliary Transport of Rosuvastatin In Vivo

    Detta är en avhandling från Uppsala : Acta Universitatis Upsaliensis

    Författare :Ebba Bergman; Uppsala universitet.; [2009]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Rosuvastatin; Biliary excretion; Transport inhibition; Pharmacokinetics; Drug-drug interactions; Hepatobiliary transport; Organic anion transporting polypeptide; OATP; Statins; Gemfibrozil; Cyclosporine; Imatinib; Rifampicin; Canalicular transport; Sinusoidal transport; Hepatic uptake; Hepatic extraction; PHARMACY Biopharmacy; FARMACI Biofarmaci; Biopharmaceutics; biofarmaci;

    Sammanfattning : In vivo studies of hepatobiliary disposition are challenging. The hepatobiliary system is complex, as its physiological localization, complex cellular structure with numerous transporters and enzymes, and the interindividual variability in protein expression and biliary flow will all affect the in vivo disposition of a drug under investigation. 3. 3. Clinical Pharmacokinetics of the Antimalarial Artemisinin Based on Saliva Sampling

    Detta är en avhandling från Uppsala : Acta Universitatis Upsaliensis

    Författare :Toufigh Gordi; Uppsala universitet.; [2001]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; Artemisinin; pharmacokinetics; saliva; HPLC; pharmacodynamics; Farmaceutisk biovetenskap; PHARMACY Biopharmacy; FARMACI Biofarmaci; Biopharmaceutics; biofarmaci;

    Sammanfattning : Artemisinin is the parent compound of a novel family of antimalarials. Repetitive administrations of artemisinin to both healthy volunteers and malaria patients have been shown to result in decreased plasma concentrations of the compound, most probably due to an autoinduction of different CYP450 enzymes. 4. 4. Models for the Transfer of Drugs from the Nasal Cavity to the Central Nervous System

    Detta är en avhandling från Uppsala : Acta Universitatis Upsaliensis

    Författare :Björn Jansson; Uppsala universitet.; [2004]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biopharmacy; Administration; intranasal; Nasal mucosa; Central nervous system; Olfactory bulb; Rat; Mouse; Swine; Cattle; Diffusion chambers; culture; In vitro; Biological models; Biological transport; Dextrans; Dopamine; Gels; Polysaccharides; Autoradiography; Fluorescence microscopy; Biofarmaci; PHARMACY Biopharmacy; FARMACI Biofarmaci;

    Sammanfattning : The blood-brain barrier restricts the access of many compounds, including therapeutic agents, to the brain. Several human studies indicate that nasal administration of hydrophilic compounds, such as peptides, can bypass the blood-brain barrier. The aims of this thesis were to develop and refine models for this direct nose-to-brain transfer. LÄS MER

  5. 5. Physiologically based pharmacokinetic modeling in risk assessment - Development of Bayesian population methods

    Detta är en avhandling från Uppsala : Acta Universitatis Upsaliensis

    Författare :Fredrik Jonsson; Uppsala universitet.; [2001]
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; methyl chloride; dichloromethane; toluene; styrene; uncertainty; intra- individual variability; risk assessment; physiologically based modeling; Markov chain Monte Carlo; PBPK; Bayesian; population modeling; Farmaceutisk biovetenskap; PHARMACY Biopharmacy; FARMACI Biofarmaci; Biopharmaceutics; biofarmaci;

    Sammanfattning : In risk assessment of risk chemicals, variability in susceptibility in the population is an important aspect. The health hazard of a pollutant is related to the internal exposure to the chemical, i.e. the target dose, rather than the external exposure. LÄS MER
