  1. 1. In vitro and in silico prediction of drug-drug interactions with transport proteins

    Författare :Gustav Ahlin; Per Artursson; Christel Bergström; Johan Karlsson; Jörg König; Uppsala universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Solute carrier; SLC transporter; OCT1; Organic cation transporter 1; SLC22A1; OATP1B1; SLCO1B1; Organic anion transporting peptide 1B1; Drug transport; Active transport; Genetic polymorphism; Cell lines; Gene expression; Multivariate data analysis; OPLS; Pharmaceutics; Galenisk farmaci; Biopharmacy; Biofarmaci; galenisk farmaci; Pharmaceutics; Biopharmaceutics; biofarmaci;

    Sammanfattning : Drug transport across cells and cell membranes in the human body is crucial for the pharmacological effect of drugs. Active transport governed by transport proteins plays an important role in this process. A vast number of transport proteins with a wide tissue distribution have been identified during the last 15 years. LÄS MER

  2. 2. The Hepatobiliary Transport of Rosuvastatin In Vivo

    Författare :Ebba Bergman; Hans Lennernäs; Ulf Bondesson; Jack Cook; Uppsala universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Rosuvastatin; Biliary excretion; Transport inhibition; Pharmacokinetics; Drug-drug interactions; Hepatobiliary transport; Organic anion transporting polypeptide; OATP; Statins; Gemfibrozil; Cyclosporine; Imatinib; Rifampicin; Canalicular transport; Sinusoidal transport; Hepatic uptake; Hepatic extraction; Biopharmacy; Biofarmaci; Biopharmaceutics; biofarmaci;

    Sammanfattning : In vivo studies of hepatobiliary disposition are challenging. The hepatobiliary system is complex, as its physiological localization, complex cellular structure with numerous transporters and enzymes, and the interindividual variability in protein expression and biliary flow will all affect the in vivo disposition of a drug under investigation. 3. 3. Clinical Pharmacokinetics of the Antimalarial Artemisinin Based on Saliva Sampling

    Författare :Toufigh Gordi; Uppsala universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; Artemisinin; pharmacokinetics; saliva; HPLC; pharmacodynamics; Farmaceutisk biovetenskap; Biopharmacy; Biofarmaci; Biopharmaceutics; biofarmaci;

    Sammanfattning : Artemisinin is the parent compound of a novel family of antimalarials. Repetitive administrations of artemisinin to both healthy volunteers and malaria patients have been shown to result in decreased plasma concentrations of the compound, most probably due to an autoinduction of different CYP450 enzymes. 4. 4. Physiologically based pharmacokinetic modeling in risk assessment - Development of Bayesian population methods

    Författare :Fredrik Jonsson; Uppsala universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; methyl chloride; dichloromethane; toluene; styrene; uncertainty; intra- individual variability; risk assessment; physiologically based modeling; Markov chain Monte Carlo; PBPK; Bayesian; population modeling; Farmaceutisk biovetenskap; Biopharmacy; Biofarmaci; Biopharmaceutics; biofarmaci;

    Sammanfattning : In risk assessment of risk chemicals, variability in susceptibility in the population is an important aspect. The health hazard of a pollutant is related to the internal exposure to the chemical, i.e. the target dose, rather than the external exposure. LÄS MER

  5. 5. In vitro evaluation of formulations used in the treatment of hepatocellular carcinoma

    Författare :Emelie Ahnfelt; Hans Lennernäs; Erik Sjögren; Niklas Axén; Jesper Østergaard; Uppsala universitet; []
    Nyckelord :MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Drug delivery system; Doxorubicin; Microgel; Emulsion; Hepatocellular carcinoma; Biofarmaci; Biopharmaceutics;

    Sammanfattning : Hepatocellular carcinoma (HCC) causes ~ 600,000 deaths annually, making it the second most deadly cancer form. HCC is classified into five stages and for the intermediate HCC treatment, the two most commonly used drug delivery systems (DDSs) are lipiodol-based emulsions and drug-eluting beads. LÄS MER
