    Författare :Charles Dahlsson Leitao; Stefan Ståhl; John Löfblom; Christoph Rader; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cancer therapy; radionuclide imaging; affibody molecules; HER-receptors; prodrug; protein design; Cancerterapi; radionuklidavbildning; affibody molekyler; HER-receptorer; prodrog; proteindesign; Bioteknologi; Biotechnology;

    Sammanfattning : Proteins are remarkable molecules with diverse and specialized functions playing essential roles in most biological processes. One such function is protecting us from diseases by the action of antibodies in our immune system that can recognize and mediate the destruction of invading pathogens by binding to foreign epitopes found on non-self proteins. Sammanfattning : Affibody molecules are small (7 kDa) affinity proteins of non-immunoglobulin origin that have been generated to specifically interact with a large number of clinically important molecular targets.In this thesis, Affibody molecules have been employed as tracers for radionuclide molecular imaging of HER2- and IGF-1R-expressing tumors, paper I-IV, and for surface knock-down of EGFR, paper V. 3. 18. Affibody Molecules Targeting VEGFR2 - Two Turns Off and Four Turns On

    Författare :Rezan Güler; John Löfblom; Stefan Ståhl; Dario Neri; KTH; []
    Nyckelord :Protein engineering; affibody molecules; VEGFR2; angiogenesis; Bioteknologi; Biotechnology;

    Sammanfattning : The notion of employing proteins as drugs traces back several decades. As recombinant DNA technology emerged, it became a powerful tool for the tailoring of protein traits via genetic approaches - so-called protein engineering. LÄS MER

  4. 19. Development of Affibody molecules for radionuclide molecular imaging and therapy of cancer

    Författare :Hadis Honarvar; Vladimir Tolmachev; Raymond M. Reilly; Uppsala universitet; []
    Nyckelord :Affibody molecules; HER2; Molecular imaging; Radionuclide targeted therapy; Radionuclide molecular imaging; Labeling chemistry; Biomedical Radiation Science; Biomedicinsk strålningsvetenskap;

    Sammanfattning : Affibody molecules are a promising class of scaffold-based targeting proteins for radionuclide-based imaging and therapy of cancer. This thesis work is based on 5 original research articles (papers I-V), which focus on optimization of molecular design of HER2-binding Affibody variants for high contrast imaging of this predictive biomarker as well as development of Affibody molecules suitable for radionuclide-based targeted therapies. 5. 20. Affibody molecules targeting the epidermal growth factor receptor for tumor imaging applications

    Författare :Mikaela Friedman; Stefan Ståhl; Fredrik Nilsson; James S. Huston; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Affibody; affinity maturation; phage display selection; EGFR; molecular imaging; protein engineering; tumor targeting; Bioengineering; Bioteknik;

    Sammanfattning : Tumor targeting and molecular imaging of protein markers specific for or overexpressed in tumors can add useful information in deciding upon treatment and assessing the response to treatment for a cancer patient. The epidermal growth factor receptor (EGFR) is one such tumor-associated receptor, which expression is abnormal or upregulated in various cancers and associated with a poor patient prognosis. LÄS MER
