1. Development of Affibody molecules for radionuclide molecular imaging and therapy of cancer
Sammanfattning : Affibody molecules are a promising class of scaffold-based targeting proteins for radionuclide-based imaging and therapy of cancer. This thesis work is based on 5 original research articles (papers I-V), which focus on optimization of molecular design of HER2-binding Affibody variants for high contrast imaging of this predictive biomarker as well as development of Affibody molecules suitable for radionuclide-based targeted therapies. 2. Engineering of Affibody molecules for Radionuclide Molecular Imaging and Intracellular Targeting
Sammanfattning : Affibody molecules are small (7 kDa) affinity proteins of non-immunoglobulin origin that have been generated to specifically interact with a large number of clinically important molecular targets. In this thesis, Affibody molecules have been employed as tracers for radionuclide molecular imaging of HER2- and IGF-1R-expressing tumors, paper I-IV, and for surface knock-down of EGFR, paper V. 3. Preclinical Molecular Imaging using Multi-Isotope Digital Autoradiography - Techniques and Applications
Sammanfattning : Molecular imaging, both in vivo and ex vivo, is playing an increasingly important role in preclinical medical research. When using radionuclide-labeled tracers, e.g. in the development of radiopharmaceuticals for diagnostic imaging or for radionuclide therapy, quantitative in vivo imaging can be performed using emission tomography. 4. Molecular Radionuclide Imaging Using Site-specifically Labelled Recombinant Affibody Molecules : Preparation and Preclinical Evaluation
Sammanfattning : Radionuclide molecular imaging is an emerging multidisciplinary technique that is used in modern medicine to visualise diseases at cellular and molecular levels. This thesis is based on five papers (I-V) and focuses on the development of site-specific radiolabelled recombinant anti-HER2 Affibody molecules and preclinical evaluations in vitro and in vivo of the labelled conjugates. 5. On the Design of Affibody Molecules for Radiolabeling and In Vivo Molecular Imaging
Sammanfattning : Affibody molecules have lately shown great potential as tools for in vivo molecular imaging. These small, 3-helical bundles, with their highly stable protein scaffold, are well suited for the often harsh conditions of radiolabeling. LÄS MER
