16. Sensitive Forensic DNA Analysis : Application of Pyrosequencing and Real-time PCR Quantification
Sammanfattning : The field of forensic genetics is growing fast and the development and optimisation of more sensitive, faster and more discriminating forensic DNA analysis methods is highly important. In this thesis, an evaluation of the use of novel DNA technologies and the development of specific applications for use in forensic casework investigations are presented. LÄS MER
17. Association of Amphiphiles and Polyelectrolytes with DNA
Sammanfattning : DNA- cationic amphiphile and DNA-cationic polymer systems were studied with respect to the intermolecular interactions, as well as structure and stability of formed complexes. The thesis is divided in two parts: one section is a summary and the other section contains five papers. LÄS MER
18. Controlling DNA compaction and the interaction with model biomembranes
Sammanfattning : The studies described in this thesis forms part of a larger collaborative project, Neonuclei, with the objective to design a module for packaging the negatively charged DNA into a ”neonucleus” with transcription competence. There are many applications that require control of DNA compaction, i.e. packaging or condensation. LÄS MER
19. DNA-Controlled Lipid-Membrane Fusion
Sammanfattning : Membrane fusion is essential for nerve-cell communication, for protein transport between cell organelles and the cell-membrane and for enabling the merger between virus and host membranes during virus infection. We have demonstrated that short DNA oligonucleotides, membrane-attached via CH in an orientation that mimics the overall zipperlike architecture of fusion-inducing proteins, induce fusion of both suspended vesicles and vesicles site-specifically tethered to SLBs. 20. Structural Adaptation of DNA to Small Ligands and Recombination Proteins
Sammanfattning : In order to contribute to the understanding of mechanisms of recognition in nucleic acid systems, the interaction of DNA with a variety of ligands, representing different cases of adaptation of the double-helix structure, was studied using flow linear dichroism (LD), circular dichroism (CD), fluorescence and absorption spectroscopic techniques. A chiral hydrophobic metal complex, [Ru(dpphen)3]2+ was examined, since this had been reported to be the most efficient molecule to discriminate DNA handedness. LÄS MER
