1. Development of benzoquinoquinoxaline derivatives as triplex-specific probes : Recognition of DNA structures at repeats sequences
Sammanfattning : Repeat sequences are associated with several human diseases, such as Friedreich’s ataxia, polycystic kidney disease and cancer. These sequences can form non-B-DNA structures, including triplex (H-DNA) DNA, and are associated with genomic instability and altered gene expression. LÄS MER
2. Structural Adaptation of DNA to Small Ligands and Recombination Proteins
Sammanfattning : In order to contribute to the understanding of mechanisms of recognition in nucleic acid systems, the interaction of DNA with a variety of ligands, representing different cases of adaptation of the double-helix structure, was studied using flow linear dichroism (LD), circular dichroism (CD), fluorescence and absorption spectroscopic techniques. A chiral hydrophobic metal complex, [Ru(dpphen)3]2+ was examined, since this had been reported to be the most efficient molecule to discriminate DNA handedness. LÄS MER
3. Fixation of Functionalized DNA Nanostructures
Sammanfattning : This thesis features results from two different studies on a fully addressable non-repetitive network on the nanometre scale, assembled by synthetic DNA molecules. A bio-inspired molecular approach, based on principles of self-assembly, has brought about a paradigm shift in nanoscience, from a top-down to a bottom-up perspective. LÄS MER
4. Bottom-up Fabrication of Functional DNA Nanostructures
Sammanfattning : This thesis demonstrates bottom-up fabrication of a fully addressable non-repetitive network on the nanometer scale, assembled by synthetic DNA molecules. Each side constitutes a unique sequence of 10 bases, i.e. 3. LÄS MER
5. Intelligent Nucleic Acid Interactions with Peptide Nucleic Acid (PNA) and Recombination Enzyme RecA
Sammanfattning : The work presented in this thesis focuses on interactions between nucleic-acids in biological systems, particularly interactions of the promising drug-candidate peptide nucleic acid (PNA), and on protein-DNA intermediates during recombination. Several experimental techniques, mainly circular dichroism and fluorescence spectroscopy, have been used to characterize the interactions of interest. LÄS MER
