1. Sequence-based predictions of membrane-protein topology, homology and insertion
Sammanfattning : Membrane proteins comprise around 20-30% of a typical proteome and play crucial roles in a wide variety of biochemical pathways. Apart from their general biological significance, membrane proteins are of particular interest to the pharmaceutical industry, being targets for more than half of all available drugs. LÄS MER
2. From protein production to genome evolution in Escherichia coli
Sammanfattning : The aim of my Ph.D. studies was to improve production yields of membrane- and secretory proteins in the widely used E. coli protein production strain BL21(DE3). LÄS MER
3. Membrane Protein Biogenesis in Escherichia coli : A proteomics approach
Sammanfattning : In the bacterium Escherichia coli all proteins are synthesized in the cytoplasm. However, at least 25% of them then need to be exported to another cellular compartment where they exert their function. E. coli has several different targeting pathways to ensure the correct localization of its proteins. LÄS MER
4. Protein targeting, translocation and insertion in Escherichia coli : Proteomic analysis of substrate-pathway relationships
Sammanfattning : Approximately 10% of the open reading frames in the genome of the Gram-negative bacterium E. coli encodes secretory proteins, and 20% encodes integral inner membrane proteins (IMPs). LÄS MER
5. Translocation of proteins into and across the bacterial and mitochondrial inner membranes
Sammanfattning : Translocons are dynamic protein complexes with the ability to respond to specific signals and to transport polypeptides between two distinct environments. The Sec-type translocons are examples of such machineries that can interconvert between a pore forming conformation that translocates proteins across the membrane, and a channel-like conformation that integrates proteins into the membrane by lateral opening.
