1. Structural studies on the integral membrane protein, ubiquinol oxidase from Escherichia coli
Sammanfattning : Heme-copper oxidases are redox-driven proton pumps that couple the reduction of molecular oxygen to water with the vectorial translocation of protons across the membrane. The proton gradient generated by heme-copper oxidases and the other members of the aerobic respiratory chain is ultimately used to drive the synthesis of ATP. LÄS MER
2. Partition chromatography of drugs on immobilized liposomes and biomembranes : A method applicable to screening in drug design
Sammanfattning : One of the main pathways for drug passage into cells is diffusion across the lipid bilayer ofthe cell membrane. A key factor in this process is the drug partitioning into the bilayer, whichcan be translated into a retention in a chromatographic system. LÄS MER
3. Multiple Functions of Glutathione Transferases : A Study on Enzymatic Function, Regulatory Role and Distribution in Mouse and Man
Sammanfattning : To cope with various endogenous toxin and xenobiotics nature has equipped the organisms with a proper protection system. Glutathione transferases (GSTs) are important components of the cellular defense against oxidative stress. These proteins appear to be suited for different tasks. LÄS MER
4. Electrochromatography and liquid chromatography : Microtechniques, instrumentation and applications
Sammanfattning : The thesis focuses on the design of efficient chromatographic and electrochromatographic microcolumns where a continuous bed is polymerized in situ in a capillary or a channel in a microchip device. In our technique, a solution of selected monomers is simply sucked or pressed into the capillary or microchip channel for subsequent polymerization. LÄS MER
5. Redesign of substrate specificity of glutathione transferase and glutathione reductase : Enzyme engineering by directed mutagenesis, phage-display selection and DNA shuffling
Sammanfattning : Human glutathione transferase (GST) A1-1 was expressed in fusion with the phage gene IIIproduct and a library of phage-displayed GST mutants was constructed by randomization of fouractive-site residues. Variant GSTs were isolated by mechanism-based phage display selection, using an immobilized transition-state analog of a nucleophilic aromatic substitution (SNAr) reaction. LÄS MER
