  1. 21. Cytotoxic Cyclotides : Structure, Activity, and Mode of Action

    Författare :Erika Svangård; Per Claeson; Lars Bohlin; Ulf Göransson; John H. Cardellina II; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacognosy; cyclotide; cytotoxic; cancer; cyclic cystine knot; Viola; Violaceae; liposome; hollow fibre; homology modelling; mass spectrometry; amphipathic; structure–activity; tumour; membrane disruption; necrosis; Farmakognosi; Pharmacognosy; Farmakognosi;

    Sammanfattning : Cyclotides are small cyclic plant proteins, and this thesis addresses their cytotoxic structure-activity properties and their mode of action on human cancer cell lines. Cyclotides were isolated from Viola odorata and Viola tricolor; three novel cyclotide sequences and two known sequences, but of new origin, were identified using mass spectrometry, amino acid analysis, and Edman degradation. 2. 22. Characterization of PAC-contaminated soil with the focus on availability, leachability and biological activities

    Författare :Monika M. Lam; Magnus Engwall; Maria Larsson; Patrik Andersson; Örebro universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Polycyclic aromatic compounds; Availability; Mobility; Soil; Risk assessment; AhR-mediated activity; ER-mediated activity;

    Sammanfattning : Current risk assessments of polycyclic aromatic compounds (PACs)-contaminated soil are often based on the 16 priority PAHs and do not consider availability of PACs in soil sufficiently. This may lead to uncertainties of the assessment, since important contaminants can be overlooked and only a small fraction of contaminants is available for the uptake for organisms. LÄS MER

  3. 23. New insights in contact allergy and drug delivery. A study of formulation effects and hapten targets in skin using two-photon fluorescence microscopy

    Författare :Carl Simonsson; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; contact allergy; cubic phases; cutaneous absorption; bromobimane; isothiocyanate; lipid vesicles; nano; pilosebaceous unit; ; two-photon microscopy;

    Sammanfattning : The skin is a remarkable barrier, protecting us from invasion of e.g. harmful microorganisms and UV-radiation. However, the skin is not adopted to resist repeated exposure to the multitude of xenobiotics introduced into modern society. LÄS MER

  4. 24. Cancer Risk Assessment of Glycidol : Evaluation of a Multiplicative Risk Model for Genotoxic Compounds

    Författare :Jenny Aasa; Margareta Törnqvist; Elaine M. Faustman; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; glycidol; 3-monochloropropane-1; 2-diol 3-MCPD ; genotoxicity; mutations; micronuclei; hemoglobin adducts; in vivo dose; multiplicative risk model; cancer risk assessment; human cancer risk; miljökemi; Environmental Chemistry;

    Sammanfattning : Humans are exposed to chemical compounds in everyday life, both from the environment and from endogenous processes. Some compounds constitute a risk for cancer development. One such compound is glycidol, which is genotoxic and an animal carcinogen. It is the model compound of this work, partly due to its presence in food. LÄS MER

  5. 25. Cardiovascular and metabolic effects of endothelins in man : an experimental and clinical study

    Författare :Astrid Ottosson-Seeberger; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :endothelin-1; big ET-1; C-terminal big ET-1 22-3 8 fragment; hemodynamic; blood flow; fractional extraction; conversion; plasma half-life; end-stage renal failure; ischemic heart disease; hemodialysis; high-flux membrane; glucose uptake; insulin sensitivity;

    Sammanfattning : Increased plasma concentrations of endothelin-1 (ET-1) and of its propeptide big endothelin-1 have been reported in diseases associated with endothelial dysfunction, cardiovascular remodelling and insulin resistance. In this thesis, we studied the hemodynamic effects of big ET-1 in healthy humans focusing on the mechanisms of the effects (study I-IV). LÄS MER
