16. Microfluidic analysis and parallel confocal detection of single molecules
Sammanfattning : The detection of single molecules using confocal spectroscopy is of major interest and tremendous improvements have been made within the last decade. The underlying basics, however, a pinhole and a single excitation volume element have remained the same as well as the technical limitations. LÄS MER
17. Amyloid β peptide: from monomer solubility to fibril structure
Sammanfattning : Alzheimer´s disease (AD) is the most common human neuro-degenerative disease characterized by the formation of cerebral plaques. Several are the amyloid-beta (Aβ) variants found in the brain of healthy individuals. LÄS MER
18. Fluorescent Probes and Protein Misfolding: Methods and Applications
Sammanfattning : Protein misfolding is a frequently occurring event in living cells and without the quality control, the consequences are disasterous. In this thesis, we studied protein misfolding in vitro by employing a variety of fluorophores, to either detect the highly ordered aggregates of intrinsically disordered proteins/peptides or slight misfolding of a natively folded protein. 19. Biophysical studies on aggregation processes and amyloid fibrils with focus on Alzheimer's disease
Sammanfattning : Alzheimer's disease (AD) is a fatal neurodegenerative disease, and is the most common form of dementia, affecting 1-2% of the population in industrialized countries. It is believed that intermediate species formed during amyloid Bpeptide (AB) polymerization play a key role in the pathogenesis of AD, but it is not known how such toxic effects are exerted. LÄS MER
20. Aβ Conformation Dependent Antibodies and Alzheimer's Disease
Sammanfattning : Soluble intermediates of the amyloid-β (Aβ) aggregation process are suggested to play a central role in the pathogenesis of Alzheimer’s disease (AD) by causing synaptic dysfunction and neuronal loss. In this thesis, soluble Aβ aggregates have been studied with a particular focus on the Aβ protofibril, which has served as the antigen for developing conformation dependent monoclonal antibodies.
