  1. 1. Molecular mechanisms of mitochondrial DNA replication

    Författare :Teresa Holmlund; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Mitochondria; mtDNA; replication; helicase; TWINKLE; POLgamma; POLgammaB; mtSSB; adPEO;

    Sammanfattning : Mitochondria are the energy producing organelles of eukaryotic cells. The organelle has its own genome, the mitochondrial DNA (mtDNA) that encodes 13 subunits of the respiratory chain (RC) complexes, two rRNAs and 22 tRNAs. LÄS MER

  2. 2. Functional and structural characterization of the human mitochondrial helicase

    Författare :Jenny Korhonen; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Mitochondria; mtDNA; replication; replisome; TWINKLE; mtSSB; POL g ; adPEO.;

    Sammanfattning : Mitochondria are the energy producing organelles of the eukaryotic cell. The human mitochondrial DNA (mtDNA) is a double stranded circular molecule of about 16 kb, usually present at 1000-10 000 copies per cell. The genome encodes 13 polypeptides involved in respiration, two ribosomal RNA s and a set of 22 transfer RNA s. LÄS MER

  3. 3. Molecular mechanisms of mammalian mitochondrial DNA replication and transcription

    Författare :Mina Pellegrini; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Mitochondrion; DNA polymerase gamma; mtSSB; TWINKLE; DNA replication; transcription; mTERF; MTERF3; MTERF2; nucleoid;

    Sammanfattning : Mitochondria are cytoplasmic organelles in eukaryotic cells, mainly devoted to the synthesis of ATP, through the Oxydative Phospshorylation System (OXPHOS). The organelle has its own genome, the mitochondrial DNA (mtDNA) that encodes for all the RNA components of the mitochondrial translation apparatus (two rRNAs and 22 tRNAs) and for 13 subunits of the OXPHOS complexes. 4. 4. DNA precursor biosynthesis-allosteric regulation and medical applications

    Författare :Reza Rofougaran; Anders Hofer; Thomas Borén; Staffan Eriksson; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; DNA biosynthesis; ribonucleotide reductase; allosteric regulation; Trypanosoma brucei; adenosine kinase; nucleoside analogs; mitochondrial DNA; TWINKLE; Biokemi; medicinsk biokemi; Medical Biochemistry;

    Sammanfattning : Ribonucleotide reductase (RNR) is a key enzyme for de novo dNTP biosynthesis. We have studied nucleotide-dependent oligomerization of the allosterically regulated mammalian RNR using a mass spectrometry–related technique called Gas-phase Electrophoretic Mobility Macromolecule Analysis (GEMMA).
