1. Studies on the transcriptional regulation of sterol 12alpha-hydroxylase (CYP8B1)
Sammanfattning : The conversion of cholesterol into bile acids is mainly via two pathways, which are initiated by CYP7A1 and CYP27A1 respectedly. CYP8B1, a microsomal P450 cytochrome, hydroxylates of the steroid nucleus at the C-12alpha position to form 7alpha, 12alpha-dihydroxy-4-cholesten-3-one, a facultative precursor of CA. LÄS MER
2. Studies of sterol 12alpha-hydroxylase (CYP8B1) : distribution and role for cholesterol homeostasis and gallstone formation
Sammanfattning : Bile acids are essential physiological molecules which carry out multiple functions such as facilitating the absorption of dietary lipids and regulating the metabolism of cholesterol and bile acids. The formation of bile represents the major route for elimination of excess cholesterol, either by conversion of cholesterol into bile acids or excretion of intact cholesterol in mixed micelles. 3. Studies on the mechanism of regulation of bile acid synthesis in humans with some aspects on genetic factors
Sammanfattning : In the present investigation we studied human bile acid synthesis and its regulation both in vivo and in vitro and also evaluated a possible influence of polymorphisms in the CYP7A1 and CYP8B1 genes on bile acid synthesis in vivo. The following observations were made: · Primary human hepatocytes cultured on matrigel under serum-free conditions produced bile acids consistent with the pattern in vivo, i. LÄS MER
4. Studies on lipoprotein metabolism and atherogenesis
Sammanfattning : Atherosclerosis is the primary cause of cardiovascular events such as angina pectoris and myocardial infarction, which together with stroke are responsible for around 50% of all deaths in the United States and Europe. Usually atherosclerosis develops over several decades and leads to a progressive narrowing of arterial vessels. LÄS MER
5. Cytochrome P450 Enzymes in Bile Acid Biosynthesis and Fatty Acid Metabolism : Studies on Members of the Porcine CYP4A and CYP8B Subfamilies
Sammanfattning : The present investigation is devoted to studies on porcine members of the cytochrome P450 4A (CYP4A) and CYP8B1 subfamilies, which are involved in bile acid biosynthesis and fatty acid metabolism. Hyocholic acid is considered to fulfil the requirements for trihydroxy bile acids in the domestic pig (Sus scrofa) in the absence of cholic acid. LÄS MER
