  1. 1. Studies on the transcriptional regulation of sterol 12alpha-hydroxylase (CYP8B1)

    Författare :Yizeng Yang; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Bile acids; CYP8B1; CYP7A1; transcription; nuclear receptor; PPAR-alpha; HNF4-alpha; FTF; SREBP; thyroid hormones.;

    Sammanfattning : The conversion of cholesterol into bile acids is mainly via two pathways, which are initiated by CYP7A1 and CYP27A1 respectedly. CYP8B1, a microsomal P450 cytochrome, hydroxylates of the steroid nucleus at the C-12alpha position to form 7alpha, 12alpha-dihydroxy-4-cholesten-3-one, a facultative precursor of CA. LÄS MER

  2. 2. Cardioprotective mechanisms by inhibition of the HMG-CoA reductase pathway and stimulation of peroxisome proliferator-activated receptors in myocardial ischaemia-reperfusion

    Författare :Aliaksandr Bulhak; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Endothelial function; endothelin-1; geranylgeranyl pyrophosphate; HMG-CoA reductase; infarct size; ischaemiareperfusion injury; myeloperoxidase; myocardium; nitric oxide; nitric oxide synthase; peroxisome proliferator-activated receptor-alpha; -gamma; PPAR-alpha; -gamma-ligands; RhoA; statins.;

    Sammanfattning : Acute myocardial ischaemia causes metabolic changes and results in a rapid decrease in the energy available to the cell. This leads to cell injury that, depending on the length of the ischaemic time, is reversible or irreversible. LÄS MER

  3. 3. Endothelial cell interactions with neutrophils : studies of adhesion and effects of lipoxygenase metabolites and antirheumatic drugs

    Författare :Mikael Heimbürger; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :adhesion; auranofin; endothelium; gold sodium thiomalate; leukotrienes; lipoxins; neutrophils; PPAR; prednisolone;

    Sammanfattning : Inflammation is a process of vital importance in the defence and elimination of microorganisms as well as in the repair of the damaged tissue. However, inadequate or excessive inflammation leads to inflammatory diseases such as rheumatoid arthritis. LÄS MER

  4. 4. The role of PPARalpha and growth hormone in hepatic lipid metaboism and atherosclerosis

    Författare :Anna Ljungberg; Göteborgs universitet; []
    Nyckelord :growth hormone; PPARalpha; atherosclerosis; lipid metabolism; fatty acids; apoB; ; ADRP; triglycerides; PGC-1; liver;

    Sammanfattning : Dyslipidemia mainly results from oversecretion of apoB-containing lipoproteins from theliver and is one of the most important risk factors for the development of atherosclerosis.Growth hormone (GH) plays a key role in the regulation of lipoprotein metabolism and thus,disturbances in GH secretion are associated with dyslipidemia and cardiovascular disease. 5. 5. Studies on nuclear receptors involved in drug metabolism

    Författare :Göran Bertilsson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : The molecular basis for drug metabolism is starting to emerge as a result of cloning and characterization of a group of nuclear receptors that respond to drugs by transcriptional regulation of key genes encoding enzymes involved in metabolism of commonly used drugs. This thesis describes the identification and functional, as well as the structural, characterization of one of these receptors, the human pregnenolone activated receptor (hPAR, NR1I2) that also has been referred to as the pregnane X receptor (PXR) or the steroid and xenobiotic receptor (SXR) by others.
