26. Studies of enzymes in mitochondrial DNA precursor synthesis : regulatory mechanisms for human thymidine kinase 2 and deoxyguanosine kinase
Sammanfattning : As important enzymes in mitochondrial nucleotide salvage pathway, thymidine kinase 2 (TK2) and deoxyguanosine kinase (dGK) are expressed constitutively in almost all cells. These two enzymes catalyze the initial rate-limiting phosphorylation of pyrimidine and purine deoxynucleosides, respectively, providing DNA precursors for mitochondrial DNA (mtDNA) replication. Sammanfattning : The overall aim of this PhD project was to gain insight into the incidence of primary immunodeficiency (PID) in southern Sweden and to optimize diagnostic and treatment measures for these patients. We estimated the occurrence rate of PID in the pediatric population of southern Sweden during a period of 4 years and described the demographic, clinical and immunological characteristics of the identified cases. In this thesis, we have investigated and characterized pathways for the processing and presentation of exogenous OVA on MHC-I ("cross-presentation") and how these were influenced by the purine nucleoside Adenosine (Ado), an immune regulator that accumulates to high concentrations at sites of inflammation and tumor growth.
