  1. 16. Macromolecules at Interfaces

    Författare :Helén Larsericsdotter; Sven Oscarsson; Jos Buijs; Willem Norde; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Chemistry; Protein; Adsorption; Immobilization; Surface; Surface Plasmon Resonance; SPR; Mass Spectrometry; MS; Differential Scanning Calorimetry; DSC; FT ICR; Lysozyme; Bovine Serum Albumin; BSA; Myoglobin; Globular; Kemi; Chemistry; Kemi;

    Sammanfattning : In this thesis, the structure and stability of globular proteins adsorbed onto nanometer-sized hydrophilic silica particles were investigated using differential scanning calorimetry (DSC), hydrogen/deuterium exchange (HDX), and mass spectrometry (MS). The adsorption process itself was characterized with fluorescence and absorption spectroscopy and surface plasmon resonance (SPR). LÄS MER

    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Hierarchical Assembly; Protein-based materials; Whey Protein Nanofibrils; β-Lactoglobulin; Recombinant Spider Silk; Microspheres; Biomedical Applications; Surface Biofunctionalization; Hierarkisk montering; Proteinbaserade material; Vassleprotein nanofibriller; β-Laktoglobulin; Rekombinant spindelsilke Mikrosfärer; Biomedicinska tillämpningar; Ytbiokonjugering; Biotechnology; Bioteknologi;

    Sammanfattning : Protein-based materials, with their unique properties of combining high strength, while maintaining elasticity, and their inherent biocompatibility, hold immense potential for various applications. In order to harness these properties, we need to understand and control how protein building blocks come together to form hierarchically structured materials. LÄS MER

  3. 18. Structural and thermodynamical basis for molecular recognition between engineered binding proteins

    Författare :Jakob Dogan; Per-Åke Nygren; John E. Ladbury; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; protein structure; induced fit; binding thermodynamics; NMR spectroscopy; protein engineering; protein-protein interactions; protein stability; calorimetry; Structural biochemistry; Strukturbiokemi;

    Sammanfattning : The structural determination of interacting proteins, both as individual proteins and in their complex, complemented by thermodynamical studies are vital in order to gain in-depth insights of the phenomena leading to the highly selective protein-protein interactions characteristic of numerous life processes. This thesis describes an investigation of the structural and thermodynamical basis for molecular recognition in two different protein-protein complexes, formed between so-called affibody proteins and their respective targets. 4. 19. Protein-Polymer Interaction and Association in Aqueous Solution

    Författare :Malin Jönsson; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Physical chemistry; Fysikalisk kemi; Makromolekylär kemi; Macromolecular chemistry; enzymologi; Proteiner; enzymology; Proteins; Aqueous two-phase systems; Monte Carlo Simulations; hydrophobic effect; surface hydrophobicity; plasmid DNA; Protein; polymer;

    Sammanfattning : The aim of this thesis has been to study protein-polymer interaction and association. Two different techniques were used; Monte Carlo simulations were combined with partitioning in polymeric aqueous two-phase systems. Monte Carlo simulations have been used to evaluate the hydrophobic interaction in electrolyte solution. LÄS MER

  5. 20. Protein engineering to explore and improve affinity ligands

    Författare :Martin Linhult; KTH; []
    Nyckelord :binding; affinity; human serum albumin HSA ; albumin-binding domain ABD ; affinity chromatography; deamidation; protein A; stabilization; Z-domain; capacity; protein G; cleaning-in-place CIP ; protein engineering; C2 receptor;

    Sammanfattning : In order to produce predictable and robust systems forprotein purification and detection, well characterized, small,folded domains descending from bacterial receptors have beenused. These bacterial receptors, staphylococcal protein A (SPA)and streptococcal protein G (SPG), possess high affinity to IgGand / or HSA. LÄS MER
