  1. 1. Benefits of Non-Linear Mixed Effect Modeling and Optimal Design : Pre-Clinical and Clinical Study Applications

    Författare :Charles Ernest II; Andrew Hooker; Mats Karlsson; Bart Ploeger; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacometrics; optimal design; nonlinear mixed effects models; population models; Pharmaceutical Science; Farmaceutisk vetenskap;

    Sammanfattning : Despite the growing promise of pharmaceutical research, inferior experimentation or interpretation of data can inhibit breakthrough molecules from finding their way out of research institutions and reaching patients. This thesis provides evidence that better characterization of pre-clinical and clinical data can be accomplished using non-linear mixed effect modeling (NLMEM) and more effective experiments can be conducted using optimal design (OD). 2. 2. Pharmacometric models to inform dose selection and study design : Applied in hemophilia and tuberculosis

    Författare :Alan Faraj; Ulrika S. H. Simonsson; Joakim Nyberg; Ron Mathot; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug development; pharmacometrics; pharmacokinetics; pharmacodynamics; non-linear mixed effect models; hemophilia; tuberculosis; dose selection; study design; pediatric extrapolation;

    Sammanfattning : While tuberculosis is a global pandemic, hemophilia is a rare disease which many have not heard of. Due to tuberculosis mainly being a problem in developing countries and hemophilia being a rare disease, they are not as heard of as other diseases such as cancer or metabolic diseases which are on the rise in Western societies. 3. 3. Pharmacometric tools to support translational drug development

    Författare :Rami Ayoun Alsoud; Ulrika S. H. Simonsson; Michael Lyons; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug development; pharmacometrics; pharmacokinetics; pharmacodynamics; non-linear mixed effect models; tuberculosis; dose selection; interspecies scaling; pediatric trials; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : The use of model-informed drug development has been shown to save significant costs and improve decision making early in the drug development process. The work in this PhD thesis aimed to employ pharmacometric tools to support translational drug development from the preclinical to the late clinical stages. LÄS MER

    Sammanfattning : ''Endotoxin and mycoplasma are Nature’s darkest secrets. If they are ever solved, Hell itself will open.'' - Lewis Thomas* Endotoxin, or lipopolysaccharides (LPS), are heterogeneous components from the cell wall of Gram-Negative bacteria and a common challenger in the field of drug discovery.
