  1. 1. Pharmacometrics to characterize innate immune response and antibacterial treatments

    Författare :Anders Thorsted; Lena E. Friberg; Elisabet I. Nielsen; Mats O. Karlsson; Gauri G. Rao; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacometrics; non-linear mixed effects modelling; modelling; pharmacokinetics; pharmacodynamics; pkpd; antibiotics; cefuroxime; piperacillin; phagocytes; lipopolysaccharide; endotoxin; porcine endotoxemia; challenge studies; sepsis; critically ill; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : The immune system protects the host against pathogens by distinguishing self from non-self. However, it is most-often disregarded in pharmacokinetic-pharmacodynamic (PKPD) characterization of (new and old) antibiotics. LÄS MER

  2. 2. Pharmacometrics for Combination Therapy in Oncology Within and Across Species Variability in Time Series and Time-to-event Data

    Författare :Marcus Baaz; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Mathematical Modeling; Pharmacometrics; Nonlinear Mixed Effects; Oncology; Combination Therapy; Radiation Therapy;

    Sammanfattning : Mathematical modeling is an integral part of the drug development process. Models are developed to describe tumor dynamics or drug concentration to answer questions such as: What concentration is required to reach the desirable treatment outcome? What drug dose and frequency should a drug prescription specify to achieve this concentration? Models are also used for simulation, reducing the need to perform additional animal trials. LÄS MER

  3. 3. Methods for handling model misspecification in pharmacometric model-based approaches for the analysis of longitudinal clinical trial data

    Författare :Estelle Chasseloup; Mats Karlsson; Andrew Hooker; Karl Brendel; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacometrics; nonlinear mixed-effects models; misspecification; type I error; power; accuracy; placebo; individual model averaging; IMA; likelihood ratio test; LRT; model averaging; covariate selection; mixture models; visual predictive checks; VPC; avatars; digital twins; virtual individual; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : Pharmacometrics model-based analyses proved to be increasingly useful in drug discovery and development. This usefulness was made evident by the formalisation of the model-integrated drug discovery and development framework (MID3). LÄS MER

  4. 4. Development, Application and Evaluation of Statistical Tools in Pharmacometric Data Analysis

    Författare :Lars Lindbom; Niclas Jonsson; Mats Karlsson; Pascal Girard; Uppsala universitet; []
    Nyckelord :Pharmaceutical biosciences; pharmacometrics; pharmacokinetics; pharmacodynamics; methodology; statistics; model evaluation; resampling methods; Farmaceutisk biovetenskap;

    Sammanfattning : Pharmacometrics uses models based on pharmacology, physiology and disease for quantitative analysis of interactions between drugs and patients. The availability of software implementing modern statistical methods is important for efficient model building and evaluation throughout pharmacometric data analyses. LÄS MER

    Författare :Kristin E Karlsson; E Niclas Jonsson; Mats O Karlsson; Anders Grahnén; Michael K Smith; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; model-based analysis; pharmacometrics; modeling; disease progression; NONMEM; SAEM; Importance sampling; repeated time-to-event; RTTCE; RCEpT; NIH stroke scale; Barthel index; internal validation; external validation; study power; study design; PHARMACY; FARMACI; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Sammanfattning : Quantitative pharmacokinetic-pharmacodynamic and disease progression models are the core of the science of pharmacometrics which has been identified as one of the strategies that can make drug development more effective. To adequately develop and utilize these models one needs to carefully consider the nature of the data, choice of appropriate estimation methods, model evaluation strategies, and, most importantly, the intended use of the model.
