1. Benefits of Non-Linear Mixed Effect Modeling and Optimal Design : Pre-Clinical and Clinical Study Applications
Sammanfattning : Despite the growing promise of pharmaceutical research, inferior experimentation or interpretation of data can inhibit breakthrough molecules from finding their way out of research institutions and reaching patients. This thesis provides evidence that better characterization of pre-clinical and clinical data can be accomplished using non-linear mixed effect modeling (NLMEM) and more effective experiments can be conducted using optimal design (OD). 2. Pharmacometric Investigations of Prediction Precision and Advances of Models for Composite Scale Data
Sammanfattning : Clinical trials are needed to evaluate new treatments. In late-stage clinical trials, failures are mostly due to lack of efficacy. Fit-for-purpose analysis methods will likely increase the success rates and advance drug development by providing higher precision to support decisions such as go/no-go, dose selection, or sample size. 3. Improved Material Models for High Strength Steel
Sammanfattning : The mechanical behaviour of the three advanced high strength steel grades, Docol 600DP, Docol 1200M and HyTens 1000, has been experimentally investigated under various types of deformation, and material models have been developed, which account for the experimentally observed behaviour. Two extensive experimental programmes have been conducted in this work. 4. Semi-mechanistic models of glucose homeostasis and disease progression in type 2 diabetes
Sammanfattning : Type 2 diabetes mellitus (T2DM) is a metabolic disorder characterized by consistently high blood glucose, resulting from a combination of insulin resistance and reduced capacity of β-cells to secret insulin. While the exact causes of T2DM is yet unknown, obesity is known to be a major risk factor as well as co-morbidity for T2DM. LÄS MER
5. Pharmacometric models to inform dose selection and study design : Applied in hemophilia and tuberculosis
Sammanfattning : While tuberculosis is a global pandemic, hemophilia is a rare disease which many have not heard of. Due to tuberculosis mainly being a problem in developing countries and hemophilia being a rare disease, they are not as heard of as other diseases such as cancer or metabolic diseases which are on the rise in Western societies.
