  1. 1. Probing the Alpha Class Glutathione Transferase Structure. Combinatorial Subunit Studies and Rational Redesign

    Författare :Ann Gustafsson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Glutathione transferases; rational redesign; enzyme; catalysis; Biokemi; Biochemistry; Biokemi; Molecular Biotechnology; molekylär bioteknik avd f jonfysik ;

    Sammanfattning : The present work concerns two Alpha class enzymes, GST A1-1 and GST A4-4, of the glutathione transferase (GST) family, which are detoxication enzymes catalyzing the conjugation of the tripeptide glutathione with various electrophiles. The GST A1-1 structure was explored for protein engineering purposes and tailored for increased activity toward alkenals. LÄS MER

  2. 2. Interplay between Pathogenic and Immune Regulatory Mechanisms in G-alpha-i2 deficient colitis

    Författare :Yu-Yuan Chiu Götlind; Göteborgs universitet; []
    Nyckelord :IBD; G-alpha-i2 colitis; Tregs; Teffs; Th17 Th1; pathogenesis immunoregulation;

    Sammanfattning : The two major forms of inflammatory bowel disease (IBD), Crohn's disease (CD) and ulcerative colitis (UC) are gastrointestinal disorders characterized by chronic and relapsing inflammation. Mice deficient in the inhibitory G protein subunit Gαi2 spontaneously develop chronic colitis and have been used as a model for IBD, with particular similarities to UC. LÄS MER

  3. 3. Activation of the spike proteins of alpha- and retroviruses

    Författare :Shang-Rung Wu; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Alphavirus; Retrovirus; Fusion protein; electron cryomicroscopy; intermediate structure;

    Sammanfattning : Enveloped viruses like human immunodeficiency virus type 1 (HIV-1) enter cells by fusing their membrane with that of the cell plasma membrane, while others, like influenzavirus and alphavirus enter via uptake into endosomes and fusion with the endosomal membrane. For this they carry spike proteins. LÄS MER

  4. 4. Redesign of Alpha Class Glutathione Transferases to Study Their Catalytic Properties

    Författare :Lisa O Nilsson; Ralf Morgenstern; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; glutathione transferase; heterologous expression; heterodimer; hydroxyalkenal; protein redesign; sequential design; dimer-interface interactions; enzyme kinetics; glutathione; dynamic C-terminus; viscosity dependence; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : A number of active site mutants of human Alpha class glutathione transferase A1-1 (hGST A1-1) were made and characterized to determine the structural determinants for alkenal activity. The choice of mutations was based on primary structure alignments of hGST A1-1 and the Alpha class enzyme with the highest alkenal activity, hGST A4-4, from three different species and crystal structure comparisons between the human enzymes. 5. 5. Regulation of exocytosis by Ca2+ and cAMP - A study on pancreatic beta- and alpha-cells

    Författare :Xiaosong Ma; Institutionen för experimentell medicinsk vetenskap; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Endocrinology; secreting systems; type-2 diabetes; SUR1; Ca2 -channel; glucagon; exocytosis; insulin; diabetology; Endokrinologi; sekretion; diabetologi;

    Sammanfattning : Type-2 diabetes is characterized by impaired insulin secretion associated with excess glucagon release. Exocytosis of insulin- or glucagon-containing granules is initiated by Ca2+-influx through voltage-dependent Ca2+-channels and is modulated by the second messengers such as cAMP. LÄS MER
