  1. 1. Rational redesign of Candida antarctica lipase B

    Författare :Anders Magnusson; Karl Hult; Bernhard Hauer; KTH; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Candida antarctica lipase B; rational redesign; secondary alcohols; substrate-assisted catalysis; S-selective; entropy; aldolase; stereospecificity pocket; oxyanion hole.; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : This thesis describes the use of rational redesign to modify the properties of the enzyme Candida antarctica lipase B. Through carefully selected single-point mutations, we were able to introduce substrate-assisted catalysis and to alter the reaction specificity. LÄS MER

    Författare :Malena A Norrgård; Bengt Mannervik; Birgitta Tomkinson; Timothy Mantle; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; protein redesign; semi-rational redesign; saturation mutagenesis; iterative saturation mutagenesis; chemical modification; Cys; Cys-X scanning; enzyme evolution; promiscuous; substrate selectivity; enantioselectivity; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Sammanfattning : Today, enzymes are extensively used for many industrial applications, this includes bulk and fine-chemical synthesis, pharmaceuticals and consumer products. Though Nature has perfected enzymes for many millions of years, they seldom reach industrial performance targets. LÄS MER

  3. 3. Probing the Alpha Class Glutathione Transferase Structure. Combinatorial Subunit Studies and Rational Redesign

    Författare :Ann Gustafsson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Glutathione transferases; rational redesign; enzyme; catalysis; Biokemi; Biochemistry; Biokemi; Molecular Biotechnology; molekylär bioteknik avd f jonfysik ;

    Sammanfattning : The present work concerns two Alpha class enzymes, GST A1-1 and GST A4-4, of the glutathione transferase (GST) family, which are detoxication enzymes catalyzing the conjugation of the tripeptide glutathione with various electrophiles. The GST A1-1 structure was explored for protein engineering purposes and tailored for increased activity toward alkenals. LÄS MER

    Författare :Olof Modén; Bengt Mannervik; Mikael Widersten; Kenneth Tew; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; allelic variants; azathioprine; bioactivation; chimeric mutagenesis; directed evolution; DNA shuffling; enzyme engineering; glutathione transferase; GST; lysate screening; molecular docking; multiple alignment; multivariate analysis; polymorphism; principal component analysis; prodrug; prodrug activation; protein engineering; protein redesign; reduced amino acid alphabet; saturation mutagenesis; semi-rational enzyme engineering; site-directed mutagenesis; structure-activity relationship; structure-based redesign; Biokemi; Biochemistry;

    Sammanfattning : Glutathione transferase (GST) A2-2 is the human enzyme most efficient in catalyzing azathioprine activation. Structure-function relationships were sought explaining the higher catalytic efficiency compared to other alpha class GSTs. LÄS MER

  5. 5. Exploring the Functional Plasticity of Human Glutathione Transferases : Allelic Variants, Novel Isoenzyme and Enzyme Redesign

    Författare :Ann-Sofie Johansson; Philip Board; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : Glutathione transferases (GSTs) make up a superfamily that is involved in the cellular defense against various reactive compounds by catalyzing the conjugation of glutathione to electrophilic centra. Members of this family have also been implicated in different facets of biological signaling. LÄS MER
