  1. 1. Microsomal glutathione transferase

    Författare :Ralf Morgenstern; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : A distinct glutathione transferase which can be activated up to eightfold by sulfhydryl reagents was discovered in rat liver microsomes. This enzyme was subsequently purified to apparent homogeneity in 40 % yield using column cromatography. LÄS MER

  2. 2. The fruit fly Drosophila melanogaster GSTE6 and E7; characterization, immobilization and transgenic overexpression

    Författare :Muhammad Aslam; Bengt Mannervik; Ralf Morgenstern; Stockholms universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Glutathione transferases; drosophila melanogaster; isothiocyanates; toxicity; immobilization; kinetics; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Sammanfattning : Glutathione transferases (GSTs) are multifunctional enzymes that are universally distributed in most eukaryotes and prokaryotes. They play a pivotal role in the metabolism and detoxication of numerous endogenous and exogenous electrophiles by conjugating them with ubiquitous tripeptide glutathione. LÄS MER

  3. 3. Role of Multiple Glutathione Transferases in Bioactivation of Thiopurine Prodrugs : Studies of Human Soluble Glutathione Transferases from Alpha, Kappa, Mu, Omega, Pi, Theta, and Zeta Classes

    Författare :Birgitta I. Eklund; Bengt Mannervik; Mikael Widersten; Ralf Morgenstern; Uppsala universitet; []
    Nyckelord :Biochemistry; Glutathione Transferases; Thiopurines; Prodrug; Bioactivation; Screening Method; Chemotherapy; Functional plasticity; Modulated activity; Biokemi;

    Sammanfattning : A screening method was developed for identification of catalytically active enzymes in combinatorial cDNA libraries of mutated glutathione transferase (GST) derivatives expressed in E. coli. The method is based on spraying monochlorobimane (MCB) directly over bacterial colonies growing on agar. LÄS MER

  4. 4. Tripeptidyl-Peptidase II : Structure, Function and Gene Regulation

    Författare :Ann-Christin Lindås; Birgitta Tomkinson; Mikael Widersten; Ralf Morgenstern; Uppsala universitet; []
    Nyckelord :Biochemistry; gene regulation; protein expression; enzyme kinetics; tripeptidyl-peptidase II; proteasome; intracellular protein degradation; exopeptidase; Biokemi;

    Sammanfattning : The protein degradation process is of vital importance for the cell to maintain cellular functions. An important enzyme in this process is the multimeric tripeptidyl-peptidase II (TPP II). It removes tripeptides from a free N-terminus of the substrates. LÄS MER

  5. 5. Redesign of Alpha Class Glutathione Transferases to Study Their Catalytic Properties

    Författare :Lisa O Nilsson; Ralf Morgenstern; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; glutathione transferase; heterologous expression; heterodimer; hydroxyalkenal; protein redesign; sequential design; dimer-interface interactions; enzyme kinetics; glutathione; dynamic C-terminus; viscosity dependence; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : A number of active site mutants of human Alpha class glutathione transferase A1-1 (hGST A1-1) were made and characterized to determine the structural determinants for alkenal activity. The choice of mutations was based on primary structure alignments of hGST A1-1 and the Alpha class enzyme with the highest alkenal activity, hGST A4-4, from three different species and crystal structure comparisons between the human enzymes.
