1. Redesign of Alpha Class Glutathione Transferases to Study Their Catalytic Properties
Sammanfattning : A number of active site mutants of human Alpha class glutathione transferase A1-1 (hGST A1-1) were made and characterized to determine the structural determinants for alkenal activity. The choice of mutations was based on primary structure alignments of hGST A1-1 and the Alpha class enzyme with the highest alkenal activity, hGST A4-4, from three different species and crystal structure comparisons between the human enzymes. 2. Protein-protein interactions in model systems : design, control of catalytic activity and biosensor applications
Sammanfattning : This thesis describes the design of polypeptides, unordered in the monomeric state but capable of folding into helix-loop-helix motifs and dimerise to form four-helix bundles. The goal of the design was to encode them with the capacity to form dimers highly selectively and the ability to carry out molecular functions in the folded state but not in the unordered state, and thus to establish a molecular link between recognition and function. Sammanfattning : The work presented in this thesis describes how structural information about a protein can be acquired, and how it can be used to answer scientific questions about proteins’ function, their dynamic behaviour and their interactions with other proteins or ligands.The catalytic function of the pyrimidine-degrading, drug metabolizing enzyme β-ureidopropionase (βUP) is dependent on the shift between oligomeric states. 5. Crystallographic studies of gluconate kinase
Sammanfattning : The first steps of gluconate metabolism in Escherichia coli are the transport of gluconate into the cell by a gluconate transport system, followed by phosphorylation to gluconate-6- phosphate by gluconate kinase.
