The transcription of the 47S pre-ribosomal RNA by RNA Polymerase I (RNA Pol I) machinery is the rate-limiting step of ribosome biogenesis and can accounts for more than 50% of total cellular transcription.     Sammanfattning : The environmental pollutant 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) induces thymus atrophy and immunosuppression in all animal species examined and these effects constitute a potential risk to humans. TCDD can via binding to the intracellular aryl hydrocarbon receptor (AhR), modulate transcription of a number of genes and the aim of this thesis has been to identify such genes, which may explain the mechanisms behind TCDD-mediated immunotoxicity.     Sammanfattning : In eukaryotes there are three DNA polymerases which are essential for the replication of chromosomal DNA: DNA polymerase alpha (Pol alpha), DNA polymerase delta (Pol delta) and DNA polymerase epsilon (Pol epsilon). In vitro studies of viral DNA replication showed that Pol alpha and Pol delta are sufficient for DNA replication on both leading and lagging DNA strands, thus leaving the function of Pol epsilon unknown.     Sammanfattning : Poly(A) polymerase (PAP) is the enzyme catalyzing the synthesis of the adenine tail to the 3’-end of mRNA. This A-tail is present on the majority of the primary RNA transcripts of protein-coding genes, and is important for mRNA stability, export to the cytoplasm and translation.
