    Sammanfattning : Capillary electrophoresis with mass spectrometric (CE–MS) detection offers a separation method without equal in terms of flexibility, utility, and cost efficiency. Here we demonstrate precisely this through the application of several laboratory-built CE–MS instruments for the separation of brain metabolites in non-primates, enantioselective separations of synthetic anesthetic metabolites in fractionated pony urine, application in structural proteomics workflows, and identification of exogenous alkaloid biotransformationproducts in human cerebrospinal fluid (CSF). Sammanfattning : In this thesis, methods for studying different aspects of proteomics were developed with Fourier Transform Ion Cyclotron Resonance, (FTICR), mass spectrometry. The FTICR technique provides ultra-high mass resolving power, mass accuracy at sub ppm level and sensitivity in the attomole region. LÄS MER

    Författare :Kristina Håkansson; Uppsala universitet; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Materials science; electrospray ionization; peptide sequencing; Fourier transform ion cyclotron resonance; hydrogen deuterium exchange; protein structure; Materialvetenskap; Materials science; Teknisk materialvetenskap; Molecular Biotechnology; Molekylär bioteknik;

    Sammanfattning : Mechanistic studies and applicability of several methods and techniques for studying peptides and proteins with mass spectrometry are presented. A model for the protein molecular ion formation in explosive matrix-assisted plasma desorption mass spectrometry is proposed, involving a contribution from the chemical energy release during conversion of the explosive, an enhanced protonation through collisions with products from the explosive decay, and electron scavenging by other products. Sammanfattning : Proteins are an important class of biomolecules, involved in the metabolism, regulation, structure and transport in cells. Unfortunately, many diseases are caused by protein dysfunction. When proteins perform their normal function, they may interact with each other or change their conformation. LÄS MER

    Sammanfattning : In this thesis, the structure and stability of globular proteins adsorbed onto nanometer-sized hydrophilic silica particles were investigated using differential scanning calorimetry (DSC), hydrogen/deuterium exchange (HDX), and mass spectrometry (MS). The adsorption process itself was characterized with fluorescence and absorption spectroscopy and surface plasmon resonance (SPR). LÄS MER
