  1. 11. Proximity Ligation : Transforming protein analysis into nucleic acid detection through proximity-dependent ligation of DNA sequence tagged protein-binders

    Författare :Simon Fredriksson; Larry Gold; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Molecular medicine; Proximity ligation; proteomics; SELEX; antibody; DNA microarray; Molekylärmedicin; Molecular medicine genetics and pathology ; Molekylär medicin genetik och patologi ; Molecular Medicine; molekylär medicin genetik och patologi ;

    Sammanfattning : A novel technology for protein detection, proximity ligation, has been developed along with improved methods for in situ synthesis of DNA microarrays. Proximity ligation enables a specific and quantitative transformation of proteins present in a sample into nucleic acid sequences. LÄS MER

  2. 12. Development of an Ultrasensitive Capacitive DNA-sensor: A promising tool towards microbial diagnostics

    Författare :Ally Mahadhy; Bioteknik; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Capacitive DNA-sensor; Enterobacteriaceae; mecA gene; Methicillin resistance; automated sequential injection analysis; sandwichhybridization; elevated temperature; polytyramine;

    Sammanfattning : Fast and sensitive detection of pathogenic microbial cells is a highly important task in medical diagnostics, environmental analysis and evaluation of food safety. Accordingly, the idea of microorganism identification by the recognition of specific DNA sequence using electrochemical technique is one of the leading researches in the development of diagnostic devices. 3. 13. Development of DNA-based methods for analysis of protein interactions

    Författare :Axel Klaesson; Ola Söderberg; Mats Nilsson; Cristina Al-Khalili Szigyarto; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; In situ proximity ligation assay PLA ; rolling circle amplification RCA ; Hybridization chain reaction HCR ; proxHCR; Oligonucleotide design; Protein-protein interactions; Post-translational modifications.; Farmaceutisk vetenskap; Pharmaceutical Science; Molekylär cellbiologi; Molecular Cellbiology;

    Sammanfattning : In situ proximity ligation assay (PLA) is a method for detection of protein interactions, post-translational modifications (PTMs) and individual proteins that allows information about their localization in a cell or tissue to be extracted. The method is based on oligonucleotide-conjugated antibodies (proximity probes) that upon binding of two epitopes in close proximity give rise to an amplifiable DNA circle. LÄS MER

  4. 14. Nanostructured surfaces created by the interactions of dendrimers and oppositely charged amphiphiles

    Författare :Marianna Yanez; Fysikalisk kemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; PAMAM; Surfactant; SDS; Lipid; Bilayer; Model Biomembrane; Nucleolipid; DLPA; DLPU; DNA; RNA; Neutron Reflectometry; Poly amidoamine ; Interface; Adsorption; Polyelectrolyte;

    Sammanfattning : The research described in this thesis aims to understand the interactions between cationic poly(amidoamine) (PAMAM) dendrimers and small oppositely charged amphiphiles at the solid-liquid and air-liquid interfaces in relation to the bulk solution behavior. There is high interest in PAMAM dendrimers because they are well-defined polymers with a hierarchical architecture which makes them promising materials as nanocapsules and gene vectors. LÄS MER

  5. 15. Quantitative analysis of DNA adducts formed by some epoxides of industrial importance

    Författare :Kamila Plná; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :DNA adducts; 32P-postlabelling; cancer; genotoxicity; occupational; epoxides; allyl glycidyl ether; propylene oxide; epichlorohydrin;

    Sammanfattning : The work presented herein has focused on DNA adduct formation by three industrial epoxides, allyl glycidyl ether, propylene oxide and epichlorohydrin. Epoxides, owing to the strained oxirane ring are chemically reactive and can alkylate various sites in DNA. LÄS MER
