  1. 1. Capturing transient peptide assemblies associated with Alzheimer's disease : Native mass spectrometry studies of amyloid-β oligomerization

    Författare :Nicklas Österlund; Astrid Gräslund; Leopold L. Ilag; Mikael Oliveberg; Sheena E. Radford; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Protein aggregation; amyloid-β; neurodegeneration; Alzheimer’s disease; chaperones; DnaJ; biomembranes; native mass spectrometry; Biophysics; biofysik;

    Sammanfattning : Correct folding of proteins is essential for maintaining a functional living cell. Misfolding and aggregation of proteins, where non-native intermolecular interactions form large and highly ordered amyloid aggregates with low free energy, is hence associated with multiple diseases. 2. 2. Secondary and Higher Order Structural Characterization of Peptides and Proteins by Mass Spectrometry

    Författare :Christopher Adams; Roman Zubarev; Göran Possnert; Carlito Lebrilla; Uppsala universitet; []
    Nyckelord :Engineering physics; Protein Structure; Mass Spectrometry; Electron Capture Dissociation; Tandem Mass Spectrometry; Teknisk fysik;

    Sammanfattning : The work in this thesis has demonstrated the advantages and limitations of using MS based technologies in protein and peptide structural studies. Tandem MS, specifically electron capture dissociation (ECD) have shown the ability to provide structural insights in molecules containing the slightest of all modifications (D-AA substitution). 3. 3. Exploring small heat shock protein chaperones by crosslinking mass spectrometry

    Författare :Wietske Lambert; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; cross-linking; mass spectrometry; protein-protein interactions; Hsp21; BS3; DTSSP; MALDI-TOF TOF; client protein; substrate; aggregation; unfolded protein; chaperone; Small heat shock protein; crosslinking;

    Sammanfattning : Together with other molecular chaperones, small heat shock proteins are key components of the protein quality control system, which is comprised of several hundred proteins and acts to maintain proteome homeostasis in the cell. Small heat shock proteins bind unfolding proteins at an early stage, to prevent these from further unfolding and aggregating. LÄS MER

  4. 4. Analysis of phospholipids in cellular membranes with LC and imaging mass spectrometry

    Författare :Ingela Lanekoff; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Mass spectrometry; TOF-SIMS; Phospholipid; Cells; Anammox;

    Sammanfattning : Imaging mass spectrometry enables the creation of molecule specific images from the surface of a solid sample in vacuum. To solve the issue of bringing single cells into vacuum without altering their native distribution of molecules, a freeze fracture device that fits the time of flight secondary ion mass spectrometry (TOF-SIMS) IV instrument has been developed. 5. 5. Native protein mass spectrometry of nuclear receptor-ligand and enzyme-substrate complexes

    Författare :Johan Lengqvist; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Mass spectrometry; electrospray; nuclear receptor; retinoid X receptor; fatty acid; membrane protein; glutathione transferase; glutathione;

    Sammanfattning : Nuclear receptors (NRs) are transcription factors that are activated by the binding of low molecular weight ligands. The NR superfamily includes receptors for steroid hormones, anti-inflammatory and lipid-lowering drugs as well as endogenous compounds such as bile acids and fatty acids. LÄS MER
