  1. 11. Free-energy studies of ligand-binding affinities

    Författare :Vilhelm Ekberg; Beräkningskemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; free energy perturbation; drug design; ligand-binding affinity; entropy; molecular mechanics; molecular dynamics;

    Sammanfattning : In drug discovery, it is of utmost importance to accurately calculate the free energies of binding ligands to various protein targets, such as enzymes and receptors. We have assessed and used computational tools for this aim, most of them based on molecular dynamics (MD) simulations. LÄS MER

  2. 12. Chromatographic Studies of Solute Interactions with Immobilized Red Blood Cells and Biomembranes

    Författare :Ingo Gottschalk; Irving W. Wainer; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Affinity; Binding; Biomembrane; Biotin; Chromatography; Cytochalasin B; Dissociation constant; Drug absorption; Equilibrium; Glucose; GLUT1; Immobilization; Immobilized biomembrane affinity chromatography; Immobilized liposome chromatography; Interaction; Liposome; Membrane protein; Membrane vesicle; Partitioning; Phospholipid bilayer; Proteoliposome; Quantitative; Red blood cell; Solute; Specific; Streptavidin; Wheat germ agglutinin; Biokemi; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Sammanfattning : Specific and non-specific interactions of solutes with immobilized biomembranes were studied using chromatographic methods. Liposomes, proteoliposomes and red blood cell (RBC) membrane vesicles were immobilized by a freeze-thawing procedure, whereas whole RBCs were adsorbed in the gel beds using electrostatic interaction, binding to wheat germ agglutinin (WGA) or the streptavidin-biotin interaction. 3. 13. Evolutionary and functional studies of protein H: a surface molecule of Streptococcus pyogenes

    Författare :Inga-Maria Frick; Infektionsmedicin; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intracellular transportation; adhesion; aggregation; FNIII domain; albumin-binding; M-like protein; convergent evolution; protein G; Streptococcus pyogenes; IgG-binding; Microbiology; bacteriology; virology; mycology; Mikrobiologi; bakteriologi; virologi; mykologi;

    Sammanfattning : Several bacterial species express surface proteins with affinity for the Fc part of human IgG. This thesis describes evolutionary and functional studies of protein H, an IgGFc-binding surface protein of the human pathogen Streptococcus pyogenes. LÄS MER

  4. 14. Visualization of Peripheral Pain Generating Processes and Inflammation in Musculoskeletal Tissue using [11C]-D-deprenyl PET

    Författare :Mikko Aarnio; Torsten Gordh; Jens Sörensen; Magnus Peterson; Lars Ståhle; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ankle injuries; arthritis; binding site; binding target; carbon-11; deprenyl; high-throughput screening; inflammation; monoamine oxi-dase; pain; PET; whiplash; Anestesiologi och intensivvård; Anaesthesiology and Intensive Care;

    Sammanfattning : An objective visualization and quantification of pain-generating processes in the periphery would alter pain diagnosis and represent an important paradigm shift in pain research. Positron emission tomography (PET) radioligand [11C]-D-deprenyl has shown an elevated uptake in painful inflammatory arthritis and whiplash-associated disorder. LÄS MER

  5. 15. Endonuclease II - a GIY-YIG enzyme of bacteriophage T4

    Författare :Pernilla Lagerbäck; Karin Carlson; Elisabeth Raleigh; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; GIY-YIG; EndoII; endonuclease; structure; tetramer; binding; nicking; recombination; Microbiology; Mikrobiologi;

    Sammanfattning : Endonuclease II (EndoII) of bacteriophage T4 is a GIY-YIG enzyme involved in host DNA breakdown during phage infection of E. coli. EndoII combines features of restriction endonucleases with those of homing endonucleases in that it breaks down DNA foreign to itself but recognizes a 16 bp long asymmetric and ambiguous sequence.
