11. Free-energy studies of ligand-binding affinities
Sammanfattning : In drug discovery, it is of utmost importance to accurately calculate the free energies of binding ligands to various protein targets, such as enzymes and receptors. We have assessed and used computational tools for this aim, most of them based on molecular dynamics (MD) simulations. LÄS MER
12. Chromatographic Studies of Solute Interactions with Immobilized Red Blood Cells and Biomembranes
Sammanfattning : Specific and non-specific interactions of solutes with immobilized biomembranes were studied using chromatographic methods. Liposomes, proteoliposomes and red blood cell (RBC) membrane vesicles were immobilized by a freeze-thawing procedure, whereas whole RBCs were adsorbed in the gel beds using electrostatic interaction, binding to wheat germ agglutinin (WGA) or the streptavidin-biotin interaction. 13. Evolutionary and functional studies of protein H: a surface molecule of Streptococcus pyogenes
Sammanfattning : Several bacterial species express surface proteins with affinity for the Fc part of human IgG. This thesis describes evolutionary and functional studies of protein H, an IgGFc-binding surface protein of the human pathogen Streptococcus pyogenes. LÄS MER
14. Visualization of Peripheral Pain Generating Processes and Inflammation in Musculoskeletal Tissue using [11C]-D-deprenyl PET
Sammanfattning : An objective visualization and quantification of pain-generating processes in the periphery would alter pain diagnosis and represent an important paradigm shift in pain research. Positron emission tomography (PET) radioligand [11C]-D-deprenyl has shown an elevated uptake in painful inflammatory arthritis and whiplash-associated disorder. LÄS MER
15. Endonuclease II - a GIY-YIG enzyme of bacteriophage T4
Sammanfattning : Endonuclease II (EndoII) of bacteriophage T4 is a GIY-YIG enzyme involved in host DNA breakdown during phage infection of E. coli. EndoII combines features of restriction endonucleases with those of homing endonucleases in that it breaks down DNA foreign to itself but recognizes a 16 bp long asymmetric and ambiguous sequence.
