  1. 1. RNA-mediated gene expression regulation

    Författare :Paula Petronela Groza; Francesca Aguilo; Yuri B. Schwartz; Silvia Remeseiro; Henrik Nielsen; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; RNA modifications; fibrillarin; 2 -O-methylation; translation; alternative splicing; METTL3; m6A; ZFP207; breast cancer; mouse ESCs; molekylärbiologi; Molecular Biology;

    Sammanfattning : The regulation of gene expression is a key mechanism that underlies all biological processes, from embryonic development to the onset and progression of various diseases, including cancer. A growing body of evidence places RNA molecules at the center of critical regulatory steps in gene expression. LÄS MER

  2. 2. RNA modifications and post-transcriptional control in cancer and stem cells

    Författare :Giulia Beneventi; Avdelningen för molekylär hematologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; RNA modifications; Pseudouridine; 2’-O-methylation; Splicing; Translation; non-coding RNAs; Cancer; Stem cells; Differentiation; Stress response;

    Sammanfattning : Splicing and translation are two of the key steps of post-transcriptional regulation of gene expression. Their tight regulation is essential for development, whereas their deregulation is involved in cancer pathogenesis. Nevertheless, many of the molecular mechanisms controlling these processes are still unknown. LÄS MER

  3. 3. iLocks: a novel tool for RNA assays with improved specificity

    Författare :Tomasz Krzywkowski; Mats Nilsson; Gunnar von Heijne; Weng-Onn Lui; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; RNA; miRNA; non-coding RNA; padlock probes; rolling circle amplification; invader; single cell; in situ; adenovirus; virology; diagnostics; Biochemistry; biokemi;

    Sammanfattning : The Central Dogma of molecular biology describes a framework for how genetic information is transferred in cells, placing RNA as a messenger between DNA and translated proteins. During the last years, interest in RNA research has grown tremendously due to the increasing understanding and recognition of the importance of RNA in regulation of gene expression, biochemical catalysis, and genome integrity surveillance. 4. 4. Conformationally Constrained Oligonucleotides for RNA Targeting

    Författare :Qing Li; Jyoti Chattopadhyaya; Li-He Zhang; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; conformationally constrained nucleoside; antisense oligonucleotide; RNA affinity; nuclease stability; RNase H; RNA degradation; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi;

    Sammanfattning : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. LÄS MER
