1. Conformationally Constrained Oligonucleotides for RNA Targeting
Sammanfattning : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. LÄS MER
2. Chromosome-specific adaptations of RNA stability and the roles of the roX RNAs in dosage compensation
Sammanfattning : Sex chromosomes evolved from an ancient pair of autosomes and the Y chromosome lost most of its genetic information in the process. This created two kinds of genomic imbalances: the first one between males (XY) and females (XX) and the second one between the sex chromosomes and the autosomes (X:AA). LÄS MER
3. Structural and Functional Studies of Wilms Tumour Protein - WT1; Novel Insights into Functionality of Zinc Finger Proteins
Sammanfattning : The molecular process under which message in DNA is relayed onto RNA is called transcription, and it accomplished through involvement of so-called transcription factors. Zinc finger proteins are one of the most abundant classes of regulatory proteins in eukaryotic cells where they play central roles in a variety of cellular activities. LÄS MER
4. Specific Precipitation of Biomolecules Using Synthetic Polymers
Sammanfattning : Purification is a very important stage in biotechnological processes. The traditionally used precipitation techniques are generally nonspecific and thus one or more chromatographic steps are normally required. There is thus a need for new approaches to improve existing techniques. LÄS MER
5. Targeting RNA by the Antisense Approach and a Close Look at RNA Cleavage Reaction
Sammanfattning : This thesis summarizes the results of studies on two aspects of nucleic acids. Chemically modified antisense oligonucleotides (AONs) have been evaluated with regards to their suitability for mRNA targeting in an antisense approach (Paper I – III). LÄS MER
