11. Pharmacological and molecular investigations on the mechanisms underlying resistance of human leukaemia cells to the antimetabolites methotrexate, 6-mercaptopurine and 6-thioguanine
Sammanfattning : The goal of this thesis has been to elucidate the mechanisms underlying resistance to methotrexate (MTX), 6-mercaptopurine (6-MP) and 6-thioguanine (6-TG), the antimetabolites widely used in treatment of childhood leukemia, as well as to determine the influence of 7-hydroxymethotrexate (7-OHMTX), the major metabolite of MTX, on the therapeutic action of MTX. Resistant sublines of leukemic cell lines were developed by long-term exposure to stepwise increasing concentrations of these different agents. LÄS MER
12. Role of membrane associated molecules in liver fibrosis
Sammanfattning : The aim of the study was to evaluate the role of membrane associated proteins in tissue formation and remodeling, in normal embryogenesis and in liver disease. Molecules associated with cellular membranes provide a fundamental basis for cellular interactions during development, homeostasis and disease. LÄS MER
13. Approaches to soft drug analogues of dihydrofolate reductase inhibitors : Design and synthesis
Sammanfattning : The main objective of the research described in this thesis has been the design and synthesis of inhibitors of the enzyme dihydrofolate reductase (DHFR) intended for local administration and devoid of systemic side-effects. The blocking of the enzymatic activity of DHFR is a key element in the treatment of many diseases, including cancer, bacterial and protozoal infections, and also opportunistic infections associated with AIDS (Pneumocystis carinii pneumonia, PCP). 14. Pneumococcal vaccination in inflammatory rheumatic disease. Antibody response and protection against infections
Sammanfattning : AbstractObjectives: Patients with inflammatory rheumatic diseases (IRDs) have higher risk of serious infections than healthy individuals due to their illness, comorbidities and treatments. Streptococcus pneumoniae is responsible for 30-50% of community aquired pneumonia in Europe. LÄS MER
15. Treatment of arthritis with tumour necrosis factor antagonists. Clinical, immunological and biochemical aspects
Sammanfattning : Abstract The treatment of arthritis has undergone a dramatic change since biological agents targeting specific mediators of the disease process have been introduced. Tumour necrosis factor (TNF) antagonists have been shown to reduce signs and symptoms of disease and to retard the development of tissue damage in the majority of patients. LÄS MER
