  1. 11. Affinity-, Partition- and Permeability Properties of the Human Red Blood Cell Membrane and Biomembrane Models, with Emphasis on the GLUT1 Glucose Transporter

    Författare :Christine Lagerquist Hägglund; Per Lundahl; Jørgen Vinten; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Affinity; Aromatic amino acids; Binding; Biomembrane; Biotin; Chromatography; Cytochalasin B; Dihydrocytochalasin B; Dissociation constant; Drug absorption; Ethanol; Equilibrium; Glucose; GLUT1; Immobilization; Immobilized biomembrane affinity chromatography; Immobilized liposome chromatography; Interaction; Liposome; Membrane protein; Membrane vesicle; Partitioning; Phospholipid bilayer; Proteoliposome; Quantitative; Red blood cell; Specific; Streptavidin; Tyrosine; Tryptophan; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : The human glucose transporter GLUT1 is abundant in red blood cells, the blood-brain barrier and epithelial cells, where it mediates the transport of the energy metabolite, glucose. In the present work some properties of GLUT1, including affinity binding of both substrates and inhibitors, transport rates as well as permeabilities of aromatic amino acids and drug-membrane interactions were analyzed by chromatographic methods. 2. 12. HYDROPHOBIC PEPTIDE TAGS - As tools in bioseparation and investigation of recombinant proteins

    Författare :Sara Fexby; Tillämpad biokemi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; E. coli; Biochemical technology; Biokemisk teknik; fusion protein; tyrosine; hydrophobicity; Hydrophobic interaction chromatography; aqueous two-phase system;

    Sammanfattning : This thesis describes the possibility of using hydrophobic peptide tags as tools for bioseparation and for the investigation of recombinant proteins. The hydrophobicity of a protein can easily be changed by minor modifications, such as fusions of tyrosine-containing peptides. LÄS MER

  3. 13. Expression and Mutation Analyses of Candidate Cancer Genes In Situ

    Författare :Sara Kiflemariam; Tobias Sjöblom; Fredrik Pontén; Mef Nilbert; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cancer; in situ; in situ hybridization; padlock probes; mRNA; tyrosine kinases; tyrosine phosphatases; fusion transcripts;

    Sammanfattning : Cancers display heterogeneity in genetic profiles of the individual cancer cells and in the composition of different malignant and non-malignant cell populations. Such intra-tumor heterogeneity plays a role in treatment response and the emergence of resistance to cancer therapies. LÄS MER

  4. 14. Regulation of Proton Coupled Electron Transfer from Amino Acids in Artificial Model Systems: A Mechanistic Study

    Författare :Martin Sjödin; Leif Hammarström; Jan Davidsson; G. Daniel Nocera; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Physical chemistry; Proton Coupled Electron Transfer; Tyrosine; Tryptophan; Electron Transfer; Hydrogen bond; Photosystem II; Proton transfer; Radical protein; Fysikalisk kemi; Physical chemistry; Fysikalisk kemi;

    Sammanfattning : Amino acid radicals are key redox intermediates in several natural enzymes including Cytochrome c peroxidase, DNA photolyase, ribonucletide reductase, cytochrome c oxidase and photosystem II. Electron transfer from amino acids is often coupled to deprotonation and this thesis concerns the coupling of electron transfer from tyrosine and tryptophan to trisbipyridineruthenium(III) with deprotonation in model complexes. 5. 15. Development and characterisation of protecting groups that enhance the solubility of synthetic peptides : Methods to improve the aqueous solubility of hydrophobic peptides

    Författare :Karolina Wahlström; Anders Undén; Fernando Albericio; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Solid phase peptide synthesis; Fmoc chemistry; solubility problems; tryptophan; tyrosine; Nmec; Nmbu; Bioorganic chemistry; Bioorganisk kemi; Neurochemistry and Molecular Neurobiology; neurokemi med molekylär neurobiologi;

    Sammanfattning : Chemical synthesis of peptides is generally performed by solid phase peptide synthesis (SPPS). Although SPPS is a rapid and convenient method to prepare peptides, the major methodological problem in SPPS is related to solvation/solubility. LÄS MER
