  1. 1. Substrate specificity of protein kinases studied with synthetic peptides

    Författare :Reet Toomik; Uppsala universitet; []
    Nyckelord :Medical sciences; protein kinase A; protein kinase C; substrate; peptide; specificity; stereospecificity; assay; SPOTS peptides; MEDICIN OCH VÅRD; MEDICINE; MEDICIN; Medical and Physiological Chemistry; medicinsk och fysiologisk kemi;

    Sammanfattning : This thesis concentrates on the substrate specificities of protein kinases A and C (PKA, PKC). Synthetic peptides are used for that purpose, both in soluble and cellulose membrane-bound form. The, stereospecificity of both protein kinases was studied with D-amino acid containing synthetic peptides. LÄS MER

  2. 2. Exploiting enzyme promiscuity for rational design

    Författare :Cecilia Branneby; Karl Hult; Alex Zaks; KTH; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Candida antarctica lipase B; alanine racemase; substrate specificity; reaction specificity; enzyme catalytic promiscuity; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : Enzymes are today well recognized in various industrial applications, being an important component in detergents, and catalysts in the production of agrochemicals, foods, pharmaceuticals, and fine chemicals. Their large use is mainly due to their high selectivity and environmental advantage, compared to traditional catalysts. LÄS MER

  3. 3. Lipase Specificity and Selectivity : Engineering, Kinetics and Applied Catalysis

    Författare :Mohamad Takwa; Karl Hult; Bernhard Hauer; KTH; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Biokemi;

    Sammanfattning : The specificity and selectivity of the enzyme Candida antarctica lipase B (CALB) were studiedfor several substrates and applications.With help of molecular modeling, the active site of CALB was redesigned for the ring openingpolymerization of D,D‐lactide. LÄS MER

  4. 4. Enzyme substrate solvent interactions : a case study on serine hydrolases

    Författare :Linda Fransson; Karl Hult; Jürgen Pleiss; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; lipase; esterase; specificity; Bioengineering; Bioteknik;

    Sammanfattning : Reaction rates and selectivities were measured for transacylation of fatty acid esters in solvents catalysed by Candida antarctica lipase B and by cutinase from Humicola insolens. With these enzymes classical water-based enzymology can be expanded to many different solvents allowing large variations in interaction energies between the enzymes, the substrates and the surrounding. 5. 5. In vitro contracture testing for the diagnosis of malignant hyperthermia susceptibility

    Författare :Gunilla Islander; Anestesiologi och intensivvård; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intensive care; Anestesiologi; Anaesthesiology; specificity; IVCT; Malignant hyperthermia susceptibility; skeletal muscle; inheritance pattern; sensitivity; reproducibility; intensivvård;

    Sammanfattning : Malignant hyperthermia (MH) susceptibility is a rare inherited muscular disorder, which may cause a lethal increase in metabolism during anaesthesia with certain commonly used anaesthetic. Susceptible individuals are without any signs or symptoms. It is important to identify individuals at risk, since the MH reaction is preventable. LÄS MER
