11. Quantum Chemical Studies of Chemotherapeutic Drug Cisplatin : Activation and Binding to DNA
Sammanfattning : The serendipitous discovery of the potent cytotoxic properties of cisplatin brought about a revolution in the treatment of certain types of cancer, but almost fifty years later, there still remain unknown areas in the chemistry of cisplatin. There are questions regarding which form of the drug reaches its DNA target, or why certain DNA sequences are more preferred than others for reaction with cisplatin. LÄS MER
12. Allosteric Regulation and Radical Transfer in Ribonucleotide Reductase
Sammanfattning : The stability of biological life over time requires that the integrity of the genetic material of an organism, the genome, be maintained as it is passed on from one generation to the next. All known cellular life has DNA-based genomes, which are duplicated before each cell division in a process called replication. LÄS MER
13. The structural basis for the catalytic specificity of manganese lipoxygenases : 3D structure analysis of the lipoxygenase of Magnaporthe oryzae
Sammanfattning : Lipoxygenases (LOX) catalyze regio- and stereospecific oxygenation of polyunsaturated fatty acids to hydroperoxides. These hydroperoxides are further metabolized to leukotrienes and lipoxins in mammals, and are involved in asthma and inflammation. LOX of animals and plants contain iron as catalytic metal (FeLOX). 14. Building blocks for polymer synthesis by enzymatic catalysis
Sammanfattning : The search for alternatives to oil-based monomers has sparked interest for scientists to focus on the use of renewable resources for energy production, for the synthesis of polymeric materials and in other areas. With the use of renewable resources, scientists face new challenges to first isolate interesting molecules and then to process them. LÄS MER
15. Redesign of substrate specificity of glutathione transferase and glutathione reductase : Enzyme engineering by directed mutagenesis, phage-display selection and DNA shuffling
Sammanfattning : Human glutathione transferase (GST) A1-1 was expressed in fusion with the phage gene IIIproduct and a library of phage-displayed GST mutants was constructed by randomization of fouractive-site residues. Variant GSTs were isolated by mechanism-based phage display selection, using an immobilized transition-state analog of a nucleophilic aromatic substitution (SNAr) reaction. LÄS MER
