  1. 1. Exploiting enzyme promiscuity for rational design

    Författare :Cecilia Branneby; Karl Hult; Alex Zaks; KTH; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Candida antarctica lipase B; alanine racemase; substrate specificity; reaction specificity; enzyme catalytic promiscuity; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : Enzymes are today well recognized in various industrial applications, being an important component in detergents, and catalysts in the production of agrochemicals, foods, pharmaceuticals, and fine chemicals. Their large use is mainly due to their high selectivity and environmental advantage, compared to traditional catalysts. LÄS MER

  2. 2. Lipase-Catalyzed Syntheses of Telechelic Polyesters

    Författare :Magnus Eriksson; Karl Hult; Mats Martinelle; Sergio Riva; KTH; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Candida antarctica lipase B; lipase-catalysis; polymerization; telechelic; functional polyesters; ring-opening polymerization; polycondensation.; Biochemistry; Biokemi;

    Sammanfattning : Telechelic polyesters have successfully been synthesized with lipase-catalyzed polymerization. The produced telechelics had a high degree of difunctionalization, high purity (requiring little or no workup) and controlled degree of polymerization. The syntheses were performed in one-pot one-step reaction systems. LÄS MER

  3. 3. Enzyme substrate solvent interactions : a case study on serine hydrolases

    Författare :Linda Fransson; Karl Hult; Jürgen Pleiss; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; lipase; esterase; specificity; Bioengineering; Bioteknik;

    Sammanfattning : Reaction rates and selectivities were measured for transacylation of fatty acid esters in solvents catalysed by Candida antarctica lipase B and by cutinase from Humicola insolens. With these enzymes classical water-based enzymology can be expanded to many different solvents allowing large variations in interaction energies between the enzymes, the substrates and the surrounding. 4. 4. Molecular modelling - understanding and prediction of enzyme selectivity

    Författare :Linda Fransson; Karl Hult; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; molecular modelling; regioselectivity; enantioselectivity; molecular dynamics; serine hydrolase; stereospecificity; Biochemistry; Biokemi;

    Sammanfattning : Molecular modelling strategies for evaluation of enzyme selectivity wereinvestigated with a focus on principles of how molecular interactionscould be evaluated to provide information about selectivity. Althoughmolecular modelling provides tools for evaluation of geometrical andenergy features of molecular systems, no general strategies for evaluationof enzyme selectivity exist. LÄS MER

  5. 5. Enzyme selectivity as a tool in analytical chemistry

    Författare :Anders Hamberg; Karl Hult; Harry Brumer; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; substrate specificity; competing substrates; enantiomeric excess analysis; library screening; ladder sequencing; carboxypeptidase Y; 2-pyridylmethylamine; MALDI; Candia antarctica lipase B; pig liver esterase; horse liver alcohol dehydrogenase; acylated cyanohydrins; Biochemistry; Biokemi;

    Sammanfattning : Enzymes are useful tools as specific analytical reagents. Two different analysis methods were developed for use in the separate fields of protein science and organic synthesis. Both methods rely on the substrate specificity of enzymes. Enzyme catalysis and substrate specificity is described and put in context with each of the two developed methods. LÄS MER
