26. Biophysical studies of cell-penetrating peptides and of the RNR inhibitor Sml1
Sammanfattning : Several short peptides, so called cell-penetrating peptides, have the capability to transport large hydrophilic cargos through the cell membrane. The objective is to use these peptides as drug carriers and thereby enhance the uptake of drugs into cells.Three different cell-penetrating peptides are characterized in this thesis. LÄS MER
27. Cell-Penetrating Penetratin Peptides - Mechanistic Studies on Uptake Pathways and DNA Delivery Efficiency
Sammanfattning : Delivery of gene-targeted drugs is limited by the inherently poor capacity of nucleic acids to overcome the membrane barrier of the cell, and development of vectors that can promote uptake is therefore crucial. Cell penetrating peptides (CPPs) have emerged as promising vector candidates due to their ability to deliver a wide range of macromolecular cargos into cells. LÄS MER
28. Cell-penetrating peptides in model membrane systems : Interaction, structure induction and membrane effects
Sammanfattning : Despite continuing advances in the development of macromolecules, including peptides, proteins, and oligonucleotides, for therapeutic purposes, the successful application of these hydrophilic molecules has so far been hampered by their inability to efficiently traverse the plasma membrane. The discovery of a class of peptides (cell-penetrating peptides, CPPs) with the ability to mediate the non-invasive and efficient import of a whole host of cargoes, both in vitro and in vivo, has provided a new means by which the problem associated with cellular delivery can be circumvented. 29. Rational design of novel cell-penetrating peptides
Sammanfattning : Although cell-penetrating peptides (CPPs) have been an active field of research for over 20 years there are still properties such as efficiency and stability that must be improved for CPPs to reach their full potential as delivery vectors. In this thesis two peptides were designed for non-covalent oligonucleotide delivery, in an attempt to overcome two major issues hampering wider use of CPPs, namely low stability and endosomal entrapment. 30. Vectorization of oligonucleotides with cell-penetrating peptides : Characterization of uptake mechanisms and cytotoxicity
Sammanfattning : The hydrophobic plasma membrane constitutes an indispensable barrier for cells in living animals. Albeit being pivotal for the maintenance of cells, the inability to cross the plasma membrane is still one of the major obstacles to overcome in order to progress current drug development. LÄS MER
