1. Cbl in Regulation of Growth Factor Receptor Endocytosis and Actin Dynamics
Sammanfattning : Proteins belonging to the Cbl family are multidomain scaffolds that participate in numerous processes, assembling signaling complexes and mediating attachment of ubiquitin to receptor and non-receptor tyrosine kinases. We characterized a novel role for Cbl and Cbl-b in ligand-dependent internalization of growth factor receptors. 2. New Electrochemical Tools to Study Exocytosis
Sammanfattning : The work described in this thesis has the focus on the development of new analytical tools to study processes related to cellular secretion (exocytosis) in cell models. Four novel techniques were developed, allowing new ways to study processes related to exocytosis, and gain previously unattainable knowledge. LÄS MER
3. Cell-Penetrating Penetratin Peptides - Mechanistic Studies on Uptake Pathways and DNA Delivery Efficiency
Sammanfattning : Delivery of gene-targeted drugs is limited by the inherently poor capacity of nucleic acids to overcome the membrane barrier of the cell, and development of vectors that can promote uptake is therefore crucial. Cell penetrating peptides (CPPs) have emerged as promising vector candidates due to their ability to deliver a wide range of macromolecular cargos into cells. LÄS MER
4. Molecular mechanisms controlling dynamin recruitment to sites of endocytosis by SH3-domain containing proteins
Sammanfattning : Endocytosis is a general cellular mechanism, which regulates a number of important events, including recycling of synaptic vesicles. Clathrin-mediated endocytosis is the best-characterized endocytic pathway in the cell. This internalization process involves the coat protein clathrin, the adaptor protein AP2 and a number of accessory proteins. LÄS MER
5. Cytoskeletal mechanisms in synaptic vesicle recycling
Sammanfattning : This thesis explores cytoskeletal mechanisms involved in the recycling of synaptic vesicles. As an initial step it characterizes synapses formed by giant reticulospinal axons in lamprey and defines their use as a model system. The most common type of reticulospinal synapse, "simple synapse", appeared to be most suitable as an experimental model. LÄS MER
