  1. 11. Expression of Manganese Lipoxygenase and Site-Directed Mutagenesis of Catalytically Important Amino Acids : Studies on Fatty Acid Dioxygenases

    Författare :Mirela Cristea; Ernst H. Oliw; Chao Su; Olof Rådmark; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical pharmacology; Dioxygenase; Gaeumannomyces graminis; lipoxygenase; Magnaporthe grisea; oxylipin; Pichia pastoris; hydroperoxide isomerase; polyunsaturated fatty acids; Farmaceutisk farmakologi; Pharmaceutical pharmacology; Farmaceutisk farmakologi;

    Sammanfattning : Polyunsaturated fatty acids can be bioactivated by two families of dioxygenases, which either contain non-heme iron (lipoxygenases) or heme (cyclooxygenases, linoleate diol synthases and α-dioxygenases).Lipoxygenases and their products play important roles in the pathophysiology of plants and fungi. 2. 12. Structural / functional studies on human alkaline phosphatases

    Författare :Alexey Kozlenkov; Jose-Luis Millan; Hans Jörnvall; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biomedicine; mutagenesis; enzyme kinetics; computer modeling; crystallography; natural substrates; uncompetitive inhibition; isozymes; gene families; Biomedicin; Microbiology; Mikrobiologi; medicinsk genetik; Medical Genetics;

    Sammanfattning : The recent elucidation of the three-dimensional structure of human placental alkaline phosphatase (PLAP) has enabled me to perform structural studies aimed at characterizing the properties of human PLAP and tissue-nonspecific AP (TNAP) as paradigms for mammalian APs in general, using site-directed mutagenesis, protein expression, kinetic analysis and computer modeling. In Paper I, we found that a single critical E429G substitution explains the difference in stability and kinetics between the common allelic variants of PLAP and the D allozyme. LÄS MER

  3. 13. Functional mapping and in vivo metabolism of the monoclonal antibody TS1 and its single-chain fragment : Its interaction with the antigen and the anti-idiotype

    Författare :Patrik Holm; Birgitta Sundström; Lars Gedda; Karlstads universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; single-chain antibody; functional mapping; site-directed mutagenesis; immune complex; metabolism; Molecular biology; Molekylärbiologi; Biomedical Laboratory Science; Biomedicinsk laboratorievetenskap;

    Sammanfattning : Antibodies are proteins capable of specific interactions to a wide range of molecules. These interactions are facilitated by the complementary determining regions (CDR).Carcinomas are the most common of human cancers and they release significant amount of cytokeratins (CK) in the necrotic areas of the tumors. 4. 14. Computational Modelling of Ligand Complexes with G-Protein Coupled Receptors, Ion Channels and Enzymes

    Författare :Lars Boukharta; Johan Åqvist; Christopher A. Reynolds; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; computer simulations; molecular dynamics; ligand binding; free energy perturbation; linear interaction energy; binding free energy; homology modelling; structure prediction; alanine scanning; site-directed mutagenesis; hERG; GPCR; neuropeptide Y; HIV-1 reverse transcriptase; integron integrase; Molecular Biotechnology; Molekylär bioteknik;

    Sammanfattning : Accurate predictions of binding free energies from computer simulations are an invaluable resource for understanding biochemical processes and drug action. The primary aim of the work described in the thesis was to predict and understand ligand binding to several proteins of major pharmaceutical importance using computational methods. LÄS MER

  5. 15. Thermostable Phytase from a Bacillus sp.: Heterologous Production, Mutation, Characterization and Assay Development

    Författare :Tran Thi Thuy; Bioteknik; []
    Nyckelord :ß-propeller phytase; recombinant protein; site-directed mutagenesis; phytate;

    Sammanfattning : Phytase is an important enzyme in the food/feed industry. It catalyzes the hydrolysis of phytate, an anti-nutrient compound present in cereals and grains, to release orthophosphate and myo-inositol-6-phosphate with lower degrees of phosphorylation. LÄS MER
