  1. 16. Exploring the Functional Plasticity of Human Glutathione Transferases : Allelic Variants, Novel Isoenzyme and Enzyme Redesign

    Författare :Ann-Sofie Johansson; Philip Board; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : Glutathione transferases (GSTs) make up a superfamily that is involved in the cellular defense against various reactive compounds by catalyzing the conjugation of glutathione to electrophilic centra. Members of this family have also been implicated in different facets of biological signaling. LÄS MER

  2. 17. Chromatographic characterization of the human red cell glucose transporter Glut1 in the cells, in membrane vesicles and in proteoliposomes

    Författare :Andreas Lundqvist; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : The human glucose transporter Glut1 is used by many cells, especially red blood cells andbrain cell, to take up glucose and other necessary metabolites. Gentle chromatographic methods can be applied for analyses of Glut1 activities in lipid environments similar to the native one. LÄS MER

  3. 18. Redesign of Alpha Class Glutathione Transferases to Study Their Catalytic Properties

    Författare :Lisa O Nilsson; Ralf Morgenstern; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; glutathione transferase; heterologous expression; heterodimer; hydroxyalkenal; protein redesign; sequential design; dimer-interface interactions; enzyme kinetics; glutathione; dynamic C-terminus; viscosity dependence; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : A number of active site mutants of human Alpha class glutathione transferase A1-1 (hGST A1-1) were made and characterized to determine the structural determinants for alkenal activity. The choice of mutations was based on primary structure alignments of hGST A1-1 and the Alpha class enzyme with the highest alkenal activity, hGST A4-4, from three different species and crystal structure comparisons between the human enzymes. 4. 19. Modeling of enzyme kinetics on polymeric substrates by simulation : Enzymatic hydrolysis of cellulose

    Författare :Veljo Sild; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : Biopolymers are an essential constituent of living matter. To develop new products and processes based on enzyme production and modification and hydrolysis of biopolymers, enzymes with well understood mechanism are required. It is impossible to derive a universal analytical model for every particular enzyme/polymer system. LÄS MER

  5. 20. Structural Plasticity and Function in Cytochrome cd1 Nitrite Reductase

    Författare :Tove Sjögren; Christian Cambillau; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; cytochrome cd1; nitrite reductase; oxidase; redox protein; electron transfer; proton transfer; X-ray crystallography; Biokemi; Biochemistry; Biokemi; biokemi; Biochemistry;

    Sammanfattning : Cytochrome cd1 nitrite reductase is a bifunctional enzyme, which catalyses the one-electron reduction of nitrite to nitric oxide, and the four-electron reduction of oxygen to water. The latter is a cytochrome oxidase reaction. Both reactions occur on the d1 haem iron of the enzyme. LÄS MER
