    Sammanfattning : Enzymes as biological catalysts are immobilized in porous materials to improve the enzyme activity and simplify their purification from the reaction media in biocatalytic applications. Mesoporous silica (MPS) particles are used as a promising support material for the immobilization of enzymes. LÄS MER

    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; catalytic mechanisms; terpene cyclase; triterpene cyclase; solvent dynamics; protein hydration; thermodynamics; quantum tunneling; polycyclization; natural compounds; ? ?-hydrolase; esterase; amidase; enzyme engineering; biocatalysis; Bioteknologi; Biotechnology;

    Sammanfattning : Enzymes enable life by promoting chemical reactions that govern the metabolism of all living organisms. As green catalysts, they have been extensively used in industry. However, to reach their full potential, engineering is often required, which can benefit from a detailed understanding of the underlying reaction mechanism. LÄS MER

  3. 3. Molecular Simulation of Enzyme Catalysis and Inhibition

    Författare :Sinisa Bjelic; Johan Åqvist; Adrian J. Mulholland; Uppsala universitet; []
    Nyckelord :Theoretical chemistry; enzyme catalysis; enzyme inhibition; computer simulations; molecular dynamics; empirical valence bond method; structure-based inhibitor design; Teoretisk kemi;

    Sammanfattning : The reaction mechanisms for the hemoglobin degrading enzymes in the Plasmodium falciparum malaria parasite, plasmepsin II (Plm II) and histo-aspartic protease (HAP), have been analyzed by molecular simulations. The reaction free energy profiles, calculated by the empirical valence bond (EVB) method in combination with molecular dynamics (MD) and free energy perturbation (FEP) simulations are in good agreement with experimental data. Författare :Pornrapee Vongvilai; Olof Ramström; Benjamin Miller; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; dynamic covalent kinetic combinatorial resolution; Self-screening; Transesterification; Amidation; Enzyme catalysis; Nitroaldol reaction; Secondary alcohols; Strecker reaction; Aminonitriles; Racemase; Enzyme catalytic promiscuity; Chemistry; Kemi; Organic chemistry; Organisk kemi; Bioorganic chemistry; Bioorganisk kemi;

    Sammanfattning : Combined thermodynamic/kinetic events amount to a kinetically controlled Dynamic Combinatorial Resolution (DCR) process, where the lability of themolecules/aggregates are used to generate dynamics, and the species experiencing the lowest activation energy is selected via kinetic process. Bothinter- and intramolecular processes can be performed using this concept,resulting in complete resolution and associated amplification of the selected species. LÄS MER

    Sammanfattning : The research summarized in this thesis focuses on directed evolution and enzyme mechanism studies of two aldolases: 2-deoxyribose-5-phosphate aldolase (DERA) and fructose-6-phosphate aldolase (FSA).Aldolases are nature’s own catalysts for one of the most fundamental reactions in organic chemistry: the formation of new carbon-carbon bonds.
