  1. 1. Development and application of ultra-sensitive fluorescence spectroscopy and microscopy for biomolecular interaction studies

    Författare :Lei Xu; Jerker Widengren; Gerhard Schütz; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; single molecule spectroscopy; fluorescence correlation spectroscopy; stimulated emission microscopy; cancer; biomolecular interaction; co-localization; Biological Physics; Biologisk fysik;

    Sammanfattning : This thesis describes the development of sensitive and high-resolution fluorescence spectroscopic and microscopic techniques and their application to probe biomolecules and their interactions in solution, lipid membrane model systems and in cells. Paper I-IV are largely focused on methodological developments. LÄS MER

  2. 2. The physiological processing of Alzheimer-associated amyloid beta precursor protein in human and animal-derived neuronal models

    Författare :Tugce Munise Satir; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer’s disease; APP; ; human iPSCs; cortical neurons; BACE1; PSEN1; neuronal activity; neuronal differentiation;

    Sammanfattning : Alzheimer’s disease (AD) is characterized by cognitive impairment due to the loss of structure and/or function of neurons, and amyloid plaques composed of aggregated-amyloid beta (Aβ) peptides, primarily species ending at the amino acid 42 (Aβ42), are one of the major neuropathological hallmarks of AD. Aβ peptides of different lengths are produced by sequential cleavage of amyloid beta precursor protein (APP) by α-, β- and γ- secretases. LÄS MER

  3. 3. Chromosome synapsis and recombination in yeast meiosis

    Författare :Hasanuzzaman Bhuiyan; Karin Schmekel; Harry Scherthan; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; meiosis; recombination; synapsis; yeast; Molecular biology; Molekylärbiologi;

    Sammanfattning : Meiosis is a cell division process that produces haploid gametes from diploid cells. Several important meiotic events take place during prophase of meiosis I, most important being homologous chromosome pairing, meiotic recombination and formation of the synaptonemal complex (SC). 4. 4. Pathophysiological effects of alpha-synuclein on SNARE complex proteins in models of alpha-synucleinopathies

    Författare :Emma Brolin; Joakim Bergström; Anna Erlandsson; Martin Ingelsson; Arianna Bellucci; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alpha-synuclein; Parkinson’s disease; synaptosome; SNARE complex; proximity ligation assay; extracellular vesicles;

    Sammanfattning : Accumulation and spread of alpha-synuclein (α-syn) aggregates are central to the disease pathogenesis of Parkinson’s disease and dementia with Lewy bodies, collectively known as α-synucleinopathies. Native α-syn is a monomeric presynaptic protein that can act as a molecular chaperone for the soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) complex assembly. LÄS MER

  5. 5. Proximity Ligation as a Universal Protein Detection Tool

    Författare :Mats Gullberg; Charles Cantor; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Molecular medicine; Proximity ligation; proteomics; aptamer; antibody; molecular beacon; rolling circle amplification; Molekylärmedicin; Molecular medicine genetics and pathology ; Molekylär medicin genetik och patologi ; Molekylär medicin; Molecular Medicine;

    Sammanfattning : Among the great challenges in biology are the precise quantification of specific sets of proteins and analyses of their patterns of interaction on a much larger scale than is possible today.This thesis presents a novel protein detection technique - proximity ligation - and reports the development and application of a nucleic acid amplification technique, RCA.
