  1. 6. Flash Diffractive Imaging in Three Dimensions

    Författare :Tomas Ekeberg; Janos Hajdu; Keith Nugent; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; X-ray; diffraction; mimivirus; three dimensional; phase retrieval; EMC; manifold embedding; CXI; FEL; free-electron laser; single particle; Physics with specialization in Biophysics; Fysik med inriktning mot biofysik;

    Sammanfattning : During the last years we have seen the birth of free-electron lasers, a new type of light source ten billion times brighter than syncrotrons and able to produce pulses only a few femtoseconds long. One of the main motivations for building these multi-million dollar machines was the prospect of imaging biological samples such as proteins and viruses in 3D without the need for crystallization or staining. 2. 7. Coherent Diffractive Imaging with X-ray Lasers

    Författare :Max Felix Hantke; Filipe R. N. C. Maia; Janos Hajdu; Martin Svenda; Edgar Weckert; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; coherent diffractive X-ray imaging; lensless imaging; coherent X-ray diffractive imaging; flash diffractive imaging; single particle imaging; aerosol injection; electrospray injection; substrate-free sample delivery; carboxysome; phase retrieval; X-ray diffraction software; X-ray free-electron laser; XFEL; FEL; CXI; CDI; CXDI; FXI;

    Sammanfattning : The newly emerging technology of X-ray free-electron lasers (XFELs) has the potential to revolutionise molecular imaging. XFELs generate very intense X-ray pulses and predictions suggest that they may be used for structure determination to atomic resolution even for single molecules. LÄS MER

  3. 8. Creating and Probing Extreme States of Materials : From Gases and Clusters to Biosamples and Solids

    Författare :Bianca Iwan; Nicusor Timneanu; Janos Hajdu; Stacey Sorensen; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; free-electron laser; ultrashort X-rays; non-equilibrium plasma; Coulomb explosion; isotope effect; hydrodynamic expansion; ion acceleration; high intensity lasers; ablation; time-of-flight spectroscopy; Fysik; Physics;

    Sammanfattning : Free-electron lasers provide high intensity pulses with femtosecond duration and are ideal tools in the investigation of ultrafast processes in materials. Illumination of any material with such pulses creates extreme conditions that drive the sample far from equilibrium and rapidly convert it into high temperature plasma. LÄS MER

  4. 9. Evasion and Attack: Structural Studies of a Bacterial Albumin-binding Protein and of a Cephalosporin Biosynthetic Enzyme

    Författare :Sara Lejon; Janos Hajdu; Karin Valegård; Martin Noble; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; human serum albumin; GA module; albumin-binding; Finegoldia magna; deacetylcephalosporin C acetyltransferase; cephalosporin C; antibiotic; beta-lactam; biosynthesis; Acremonium chrysogenum; X-ray crystallography; Structural biology; Strukturbiologi;

    Sammanfattning : This thesis describes the crystal structures of two proteins in the context of combatting bacterial infections. The GA module is a bacterial albumin-binding domain from a surface protein expressed by pathogenic strains of the human commensal bacterium Finegoldia magna. LÄS MER

  5. 10. Towards Fast and Robust Algorithms in Flash X-ray single-particle Imaging

    Författare :Jing Liu; Stefan Engblom; Filipe Maia; Janos Hajdu; Helmut Grubmüller; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; X-ray Free Electron lasers XFELs ; 3D electron density determination; Machine learning; Image processing; High-performance computing; GPUs; Uncertainty quantification; X-ray single-particle Imaging; Flash X-ray single-particle diffraction Imaging FXI ; ; Physics with specialization in Biophysics; Fysik med inriktning mot biofysik;

    Sammanfattning : Modern X-ray Free Electron Laser (XFEL) technology provides the possibility to acquire a large number of diffraction patterns from individual biological nano-particles, including proteins, viruses, and DNA. Ideally, the collected data frames are high-quality single-particle diffraction patterns. LÄS MER
